CD40 ligand on activated platelets triggers an inflammatory reaction of endothelial cells

Abstract: CD40 ligand (CD40L, CD154), a transmembrane protein structurally related to the cytokine TNF-alpha, was originally identified on stimulated CD4+ T cells, and later on stimulated mast cells and basophils. Interaction of CD40L on T cells with CD40 on B cells is of paramount importance for the development and function of the humoral immune system. CD40 is not only constitutively present on B cells, but it is also found on monocytes, macrophages and endothelial cells, suggesting that CD40L has a broader function i… Show more

“…Future experiments are aimed at elucidating the mechanisms by with anti-CD40L mAb promote “tolerogenic” neutrophils. In this respect we hypothesize neutrophils scanning for platelets (35) may receive a “tolerogenic” signal following CD40L blockade in activated platelets (36) that may lead to CSF1 production and transplantation tolerance under sterile inflammatory conditions.…”

“…In the latter case, activated platelets expressing or releasing CD40L [22,24] could also interact with CD40 on duct cells, since islet cell suspensions are directly injected into the portal vein. CD40-CD40L interactions at the pancreatic duct cell level could therefore contribute to beta cell damage in Type 1 diabetes and after islet transplantation.…”

“…The ligand for CD40 (CD40L) is a type II transmembrane protein expressed on stimulated T cells but also on mast cells, basophils and platelets. A soluble form of CD40L is released from activated platelets or shed from stimulated lymphocytes [21,22,23,24]. CD40-CD154 interactions were shown to play a pivotal role in different T-cell-mediated inflammatory disorders [20,21,25,26,27] and disruption of CD40-CD40L interactions proved to be sufficient to prevent the development and progression of several autoimmune diseases [28,29] as well as allograft rejection [21,26].…”

“…A cellular cross-talk might be required to efficiently trigger IFN-α secretion by PMNs. Owing to their proinflammatory activity [18,19], platelets might be involved. Indeed, platelets activated by immune complexes were recently shown to potentiate IFN-α secretion by PDCs in SLE through a CD154-CD40 interaction [20].…”
