The present study was undertaken to investigate the comparative efficacy of captopril (CP) and allylmercaptocaptopril (AMC) supplementation following selenite exposure with respect to oxidant-antioxidant equilibrium and prevents cataract formation in rat pups. Forty Wistar-albino rat pups were equally divided into 4 groups. In Group I; only subcutaneous (s.c) saline was injected. In Group II; single dose of sodium selenite (19 µM/kg body weight, s.c) was injected on postpartum day 10. One day before the selenite challenge, the pups in Group III and IV were injected intraperitoneally a single dose of 50 mg/kg of CP and AMC respectively and repeated once daily thereafter up to 21 st day. The development of cataract was assessed weekly. The density of cataract was graded by slit-lamp biomicroscopy. On day 21, the blood was collected and lenses were removed. Total oxidant status (TOS), total antioxidant capacity (TAC), oxidative stress index (OSI) and malondialdehyde (MDA) level were determined in the lenses of the rat pups. Serum paraoxonase-1 (PON1) activity was determined. All of the lenses of rat pups in Group I remained clean. All rat pups developed dense nuclear opacity in Group II. Three out of 10 rat pups developed slight nuclear opacity in Group III while only one rat pup out of 10 in Group IV had vacuolization but not opacity. Differences among the groups were statistically significant (p<0.01). Group II lenses had higher mean OSI and MDA level than those of Group III and IV (p<0.01). Group II lenses had lower mean TAC level and serum PON1 activity than those of Group III and IV (p<0.01). A negative correlation was found between serum PON1 activity and lens MDA level (r = -0.6861; P<0.001). CP and AMC both diminished the incidence of cataract due to its protection of the antioxidant defense system but AMC was found to be more effective than CP.

The study was designed to investigate silymarine as preventive agent in selenite-induced cataract in rabbits. Eighteen rabbits were concluded and divided equally into negative control (I), positive control (II) and test (III) groups, each 6. After clinical and ophthalmoscope, bio-microscope slit lamp equipped with a digital camera and ultrasonographical examinations of rabbits' eyes, selenite sodium was administered subcutaneously on the neck in all rabbits at day 0 of the study. After induction of anesthesia, lateral recumbency and making a pore in globe 0.1ml of saline solution 0.9% in group II and 0.1ml of silymarine were injected into vitreous in group III. Any agent did not administrate in group I. Selenite sodium was injected in days third and sixth in all rabbits subcutaneously. The bio-microscopic slit lamp study showed grade one and two cataract in day 4 and 8, and grade three in day 11 in all rabbits in group I and II. In group III, three rabbits on the eighth day and four until the end of the eleventh day presented grade one cataract. Anterior lens capsule thickness was less in group III in comparison to group I and II, and posterior capsule thickness in group III was significant statistically by ultrasonography on day 20 and the anterior-posterior lens diagonal length in the experimental group comparing to the control groups was longer significantly. Selenite sodium showed cataractogenic character and silymarine as probable protective role on selenite -induced cataract in rabbit model. Aim: To investigate whether erdosteine supplementation following selenite exposure affects oxidant-antioxidant equilibrium and prevents cataract formation in rat pups.Methods: Thirty-nine Wistar-albino rat pups were divided into 3 groups. In Group 1 (n=16) only s.c saline was injected. In Group 2 (n=10) subcutaneous (s.c.) sodium selenite (30 nmol / g body weight) was injected on postpartum day 10. In Group 3 (n=13) s.c. sodium selenite (30 nmol/g body weight) were injected on postpartum day 10 and oral erdosteine (10 mg/kg body weight, daily for one week) was administered by gavage thereafter. The development of cataract was assessed weekly. The density of cataract was graded by slit-lamp biomicroscopy. On day 21, the blood was collected and lenses were removed. Oxidative stress index (OSI) and total antioxidant capacity (TAC) were determined in the lenses of the rat pups. Paraoxonase-1 (PON 1) activity was determined in the sera.Results: All of the lenses of rat pups in Group 1 remained clean. All rat pups developed dense nuclear opacity in Group 2. Eight out of 13 rat pups developed slight nuclear opacity in Group 3. Differences among the groups were statistically significant (p<0.05). Group 2 lenses had higher mean OSI level than that of Group 3 lenses (p=0.003). Group 2 lenses lower mean TAC levels than that of Group 3 (not significant). Mean PON 1 level of Group 2 was lower than that of Group 3 (p<0.05).Conclusion: Erdosteine diminishes the incidence of cataract due to its protection of the antioxidant defense system.
