Abstract: We previously reported that hydrogen sulfide (H(2)S) preconditioning (SP) produces cardioprotective effects against ischemia in rat cardiac myocytes. The present study aims to elucidate the signaling mechanisms involved in SP-induced cardioprotection by investigating the role of extracellular signal regulated kinase (ERK1/2) and phosphatidylinositol 3-kinase (PI3K)/Akt. We found that preconditioning with NaHS (a H(2)S donor) for three cycles significantly decreased myocardial infarct size and improved heart co… Show more

“…The ERK1/2 activation has been shown to be increased (42)(43)(44)(45)(46), decreased (22,23,29,(47)(48)(49) or unaltered (50,51) following exposure to H 2 S. In the present study, we provide clear evidence that the activation of ERK1/2 contributes to HG-induced injury in H9c2 cells and that exogenous H 2 S protects H9c2 cells against HG-induced injury by inhibiting ERK1/2 activation, which is supported by our previous study [Dong et al (23)]. Although the mechanisms through which ERK1/2 induces either cell death or survival and the reasons for the differential effects induced by exogenous H 2 S on the activation of ERK1/2 are unclear, it is generally accepted that the duration and magnitude of ERK1/2 activation may be an important factor in determining cell survival or apoptosis (52).…”

“…The activation of Akt, which is downstream of PI3K, may ameliorate I/R injury [13] . The PI3K/Akt pathway is also involved in the preconditioning of hydrogen sulfide (H 2 S), another sulfur-containing gas in the methionine metabolic pathway with effects on myocardial I/R similar to those of SO 2 [14] . The aim of the present study was to investigate whether the PI3K/Akt pathway is involved in the cardioprotective effect of SO 2 preconditioning.…”

“…Recent work from our laboratory has shown that H 2 S preconditioning produced cardiac protective effects similar to that induced by IPC (2,30). In addition, exogenous H 2 S was shown to activate several prosurvival kinases such as ERK1/2 (51, 52, 61), Akt (17), and PKC (31), which consist of the active arm of IPostC. Although the complete signaling mechanisms of H 2 S remain to be clarified, the general understanding on the cardioprotective effect of H 2 S to date has led us to hypothesize that endogenous H 2 S may contribute to the protective effect of IPostC.…”
