“Cardioplegia on the Contractile Apparatus Level”: Evaluation of a New Concept for Myocardial Preservation in Perfused Pig Hearts

Abstract: Under the given experimental conditions reversible desensitization of the contractile apparatus for calcium results in a considerable prolongation of the tolerance to cold ischemia in explanted pig hearts. The present study shows that the protective effects of BDM are not only present when isolate muscle fibres were stored (and the extracellular space is large) but also after storage of complete hearts in a solution in a solution containing BDM. Thus BDM may become a useful agent to enlarge the storage period … Show more

“…Blood withdrawn after catheterization of the inferior vena cava of the anesthetized and heparinized animal was then mixed with Krebs-Henseleit in a 1:4 proportion. 2,3 Butanedione monoxime (BDM 97%, Aldrich, Paris, France, a potent reversible inhibitor of the actin-myosin interaction (11), was added at a concentration of 30 mmol/L to the perfusate in order to inhibit motion artifacts related to contraction and to favor the homogeneity of myocardial perfusion.…”

“…У літературі багато робіт, присвячених впливу кардіоплегії на морфологічну характеристику міокарда. Причому ультраструктурні зміни вивчали здебільшого на експериментальних тваринах [2,3]. Але результати цих досліджень не можна повною мірою екстраполювати на пацієнтів кардіохірургічних клінік, тому що хворі, на відміну від тварин, поступають на операційний стіл вже із скомпрометованим міокардом.…”
