Previous studies indicated that BITC is more carcinogenic than AITC (4,5,11), and direct application of these ITCs in the present study suggested a correlation with the degree of cytotoxicity. Considering the similar cytotoxic effects on rat urinary bladder with oral administration of 0.1% BITC (manuscript in preparation) and the instillation documented here, we can surmise that mercapturic acid conjugates, such as Acetyl-B-Cys for BITC, are generated and cleaved to produce free BITC in the urine, independent of the route of application.…”

Allyl isothiocyanate (AITC) is known to be weakly carcinogenic , whereas benzyl isothiocyanate (BITC) has been suggested to exert carcinogenicity toward the rat urinary bladder. To elucidate direct toxic effects of isothiocyanate s (ITCs), BITC, AITC, or BITC-metabolites conjugated either with glutathione, cysteinylglycine , cysteine, or mercapturic acid were intravesically instilled into female F344 rats. Exposure to AITC and BITC at 2.8 mg/kg body weight, and the same mol quantity (37 l mol/kg) of BITC-metabolites was for 2 h. Nineteen hours thereafter, the animals were intravenously administered 5-bromo-2 -deoxyuridine (BrdU) and killed 1 h later. BITC caused more profound toxic damage than AITC. Among the BITC-metabolites, cytotoxicity was evident with intermediate glutathione or cysteinylglycin e conjugates, whereas the mercapturic acid, considered to be the major nal urinary metabolite, exerted little effects. BrdU labeling was essentially dependent on the degree of cytotoxic potential of each compound . Considering the previous study results demonstrating the generation of free BITC from metabolites in urine, the present results support the idea that cytotoxic activity of orally administered ITCs is derived from free forms cleaved from conjugated metabolite(s) in urine.

The human diet contains a great variety of natural mutagens and carcinogens, as well as many natural antimutagens and anticarcinogens. Many of these mutagens and carcinogens may act through the generation of oxygen radicals. Oxygen radicals may also play a major role as endogenous initiators of degenerative processes, such as DNA damage and mutation (and promotion), that may be related to cancer, heart disease, and aging. Dietary intake of natural antioxidants could be an important aspect of the body's defense mechanism against these agents. Many antioxidants are being identified as anticarcinogens. Characterizing and optimizing such defense systems may be an important part of a strategy of minimizing cancer and other age-related diseases.
