Carbohydrate and alditol analysis by high-performance anion-exchange chromatography coupled with electrochemical detection at a cobalt-modified electrode

Abstract: A cobalt oxyhydroxide film dispersed on a carbon electrode surface was characterized and proposed as an amperometric sensor for determination of alditols and carbohydrates in flowing streams. Complex mixtures of carbohydrates were separated by anion-exchange chromatography using a moderately alkaline solution as mobile phase. The cobalt modified electrode (GC-Co) was employed under a constant applied potential of 0.5 V (vs Ag/AgCl). Under these experimental conditions the detection limits (S/N=3) for all analy… Show more

“…The seed samples of each were milled into powder and (400 mg) of each were later hydrolysed with 6 N HCl for 24 hours and then derivatized with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) to produce stable derivatives. These amino acids are then analysed by reverse phase UPLC [3,8,10,11].…”

“…Based on the separation and detection method, the analytical methods used for monosaccharide composition analysis of polysaccharides include gas chromatography [17], high-performance liquid chromatography (HPLC) [18], ion chromatography (IC) [19], and capillary electrophoresis (CE) [20]. Generally, polysaccharides are difficult to detect by conventional HPLC methods using UV or fluorescence detection because of their nonchromophoric attributes [21]. Electrochemical detection (ECD) provides an alternative detection method due to its simplicity, selectivity and low cost [22].…”

“…Recent works are more oriented towards oxides of transition metals for example NiOx [11], FeOOH [12], CuO [13], MnO2 [14] , RuO2 [15], Cu2O and Co3O4 [16][17].The presence of oxidized forms provides an intense electro catalytic behaviour, being capable of oxidizing many various organic molecules such as alcohols, amino acids, sugars and nitrites [18][19][20][21][22][23].…”
