Cancer panel analysis of circulating tumor cells in patients with breast cancer

Abstract: Liquid biopsy using circulating tumor cells (CTCs) is a noninvasive and repeatable procedure, and is therefore useful for molecular assays. However, the rarity of CTCs remains a challenge. To overcome this issue, our group developed a novel technology for the isolation of CTCs on the basis of cell size difference. The present study isolated CTCs from patients with breast cancer using this method, and then used these cells for cancer gene panel analysis. Blood samples from eight patients with breast cancer were… Show more

“…CTC isolation was performed using a SMART BIOPSY SYSTEM Isolation kit (cat no. CIKW10; Cytogen, Inc., Seoul, Korea) 61 , 66 , 67 . Briefly, blood samples were incubated with 20 µg/µl of antibody cocktail (complex) from the SMART BIOPSY SYSTEM Isolation kit (Cytogen, Inc.) targets white blood cells (CD45) and red blood cells (globin), captures epithelial CTCs, which was uniquely developed by Cytogen Inc., for 20 min and were then mixed with pre-activation buffer followed by density gradient centrifugation at 400 × g for 30 min at room temperature.…”

“…Targeted NGS analysis on genomic DNA, extracted from patients’ white blood cells (WBC), was performed to exclude all germline sequence variants during the evaluation of mutational results, as previously described 32,33 . As shown in Supplementary Tables, we found several discrepancies at both intra-patient and inter-patient levels and, although in few instances we found shared gene variants in primary tumors matched with CTC samples (e.g.…”

“…Several Authors also described such heterogeneity at single-CTC level 26,29,3335,44 , but the minimal amount of DNA extracted from a single cell requires preliminary WGA for downstream molecular analyses. WGA is a time-consuming procedure that may provide technical errors such as inadequate coverage, allelic dropouts, false negative and false positive results 23,38 that may affect the analytical quality of NGS procedures 18,38 .…”

“…Even with aggressive surgery or chemoradiation therapy, microscopic cancer cells can remain. Recent work on circulating cancer cells has provided evidence of the presence of numerous microscopic cancer cells after local treatment (4). Therefore, removal of these remaining cancer cells is important for complete cure of such malignancies, a process in which host immunity plays an important role.…”
