Calibration and Application of an X-ray Image Intensifier/ Charge-Coupled Device Detector for Monochromatic Macromolecular Crystallography

Abstract: Charge-coupled device (CCD)-based X-ray detectors allow data to be collected much more quickly (--,10 times) than with current on-line imaging-plate systems. At the ESRF, X-ray image intensifier/CCD detector systems have been developed. These have great potential as fast read-out detectors for macromolecular and other forms of crystallography. They are relatively large sensitive X-ray detectors but have two inherent weaknesses: convex detection surfaces leading to spatial distortion and non-uniformity of inten… Show more

“…Buffer scattering was recorded before each sample scattering and then subtracted from the corresponding sample scattering. The integration of SAXS patterns was performed using Fit2D software (41) and data analysis using GNOM (42). Low resolution envelopes of each construction were calculated from the experimental SAXS data using ab initio procedure implemented in the DAMMIN program (43).…”

“…The remaining three B8 complexes were collected at BL4 (ESRF) (~ = 0.76 A) using an X-ray Image Intensifier Charged Couple Device (XRII)/ CCD Detector [32]. Data from the XRII/CCD Detector were first corrected for spatial distortion and non-uniformity of response over the detector system [33,34]. All diffraction data were autoindexed, integrated and corrected for Lorentz and polarization effects with the program DENZO [35] followed by scaling and merging in SCA-LEPACK [35].…”
