Calcium phosphate formation in aqueous suspensions of multilamellar liposomes

Abstract: The present study examined calcium phosphate precipitation in aqueous suspensions of multilamellar liposomes as a possible in vitro model for matrix vesicle mineralization. Liposomes were prepared by dispersing CHCl3-evaporated thin films of 7:2:1 and 7:1:1 molar mixtures of phosphatidylcholine, dicetyl phosphate, and cholesterol in aqueous solutions containing 0, 25, or 50 mM PO4 and 0 or 0.8 mM Mg. After removal of unencapsulated PO4 by gel filtration, the liposomes were suspended in 1.33 mM Ca/0.8 mM Mg sol… Show more

“…There have been previous attempts to simulate MV mineralization using multilamellar liposomal systems (Eanes et al 1984;Eanes andHailer 1985, 1987;Eanes 1989). In addition, liposomes containing various proportions of anionic and neutral phospholipids and cholesterol were used to study the ability of annexin V to facilitate Ca 2+ uptake (Kirsch et al 1997).…”

“…The proteoliposome system provides a means of reconstituting lipid vesicles that will function like MVs. There have been previous attempts to simulate MV mineralization using multilamellar liposomal systems (44,45,56,57). More recently, liposomes containing various proportions of anionic and neutral phospholipids and cholesterol were used to study the ability of annexin V to facilitate Ca 2+ uptake (26).…”

“…In studying the effect of membrane-bound acidic phospholipids on calcium phosphate (CapI) precipitation, we have used as our basic membrane model liposomes prepared from mixtures of phosphatidylcholine, dicetyl phosphate, and cholesterol, usually in the molar ratios of 7:l-2:l (Eanes et al, 1984). Although dissimilar in composition to matrix vesicle membranes, this specific mixture was chosen because the liposomes formed stable aqueous suspensions that did not readily fuse in the presence of solution Ca2+.…”

“…As detailed elsewhere (Eanes et al, 1984), the first step in the preparation of the liposomes was rotatory evaporating from chloroform the primary lipid mixture as a thin film on the inner surface of a round bottom flask. Where appropriate, the primary mixture was augmented with up to 20 mole % of one of the following acidic phospholipids: phosphatidic acid, phosphatidylglycerol, phosphatidylinositol, or phosphatidylserine.…”
