Abstract: Melanoma is an aggressive tumor with still poor therapy outcomes. δ-tocotrienol (δ-TT) is a vitamin E derivative displaying potent anti-cancer properties. Previously, we demonstrated that δ-TT triggers apoptosis in human melanoma cells. Here, we investigated whether it might also activate paraptosis, a non-canonical programmed cell death. In accordance with the main paraptotic features, δ-TT was shown to promote cytoplasmic vacuolization, associated with endoplasmic reticulum/mitochondrial dilation and protein… Show more

“…We have previously reported that δ‐TT is able to generate massive oxidative stress in prostate cancer and melanoma cells. 10 , 11 , 12 Therefore, we measured ROS formation in both IGROV‐1 and SKOV‐3 cells treated with the compound. As described in Figure 5A,B , after 12 hours of treatment (15 μg/ml) OC cells exhibited a threefold (IGROV‐1) and twofold (SKOV‐3) increase in overall cellular ROS production and a twofold (both cell lines) increase in mitochondrial ROS generation as compared to control group.…”

“… 8 , 9 We previously demonstrated that δ‐TT could induce ROS‐mediated cell death in prostate cancer and melanoma. 10 , 11 , 12 Moreover, δ‐TT has been recently reported to synergize with bevacizumab in a phase II trial conducted on chemotherapy‐refractory OC. 13 However, the molecular mechanisms underlying δ‐TT anti‐tumour activity in OC cells have not been elucidated yet.…”

“…Abnormal proinflammatory cytokine production and excess ROS generation are also associated with mitochondrial damage in vitro and in vivo . 34–36 Although 10 μm MPs cannot enter cells, they still induced the production of IL-1β, IL-6, and TNF-α, while also increasing ROS levels in ECs. Hence, 10 μm MPs cause EC mitochondrial dysfunction via increased proinflammatory cytokines and ROS production.…”

“…Additionally, δ-tocotrienol treatment also reduced mitochondrial membrane potential, oxygen consumption, and the expression of mitochondrial complex I. The mitochondrial Ca 2+ overload was thought to be mediated by IP 3 R and VDAC [116].…”
