C-Methylated Flavonoids from Cleistocalyx operculatus and Their Inhibitory Effects on Novel Influenza A (H1N1) Neuraminidase

Abstract: As part of an ongoing study focused on the discovery of anti-influenza agents from plants, four new (1-4) and 10 known (5-14) C-methylated flavonoids were isolated from a methanol extract of Cleistocalyx operculatus buds using an influenza H1N1 neuraminidase inhibition assay. Compounds 4, 7, 8, and 14, with a chalcone skeleton, showed significant inhibitory effects on the viral neuraminidases from two influenza viral strains, H1N1 and H9N2. Compound 4 showed the strongest inhibitory activity against the neuram… Show more

“…Among them, a pronounced neuroaminidase-inhibiting effect was observed for the herb extract of Elsholtzia rugulos. Some extracts from Agrimonia pilosa, 66 Echinacea purpurea, 67 and Prunus mume 68 or the multicomponent mixtures polyphenol fractions from Punica granatum 69 and secoiridoid glucosides from Ligustrum lucidum 70 have shown a significant reduction of virus-induced cytopathic effects and in general antiviral or anti-influenza activity.…”

“…The neuraminidase inhibition assay was performed as previously described with several modifications (Dao et al, 2010). Briefly, influenza H1N1 or H9N2 viruses were injected into MDCK cells in large-scale using DMEM containing trypsin (0.15 mg/mL) and bovine serum albumin (5 mg/mL) (BSA; Sigma-Aldrich Co., St Louis, MO, USA).…”

“…5,7-Dihydroxy-6,8-dimethylflavanone (2) has been found previously in a range of plants, including pōhutukawa (Metrosideros excelsa, Myrtaceae) (Mustafa et al 2005). It has been reported to have anti-bacterial (Massaro et al 2014), antimalarial (Boonphong et al 2007;Joseph et al 2007) and antiviral (Dao et al 2010) activities.…”
