Abstract: Tactics to selectively enhance cancer radioresponse are of great interest. Cancer cells actively elaborate and remodel their extracellular matrix (ECM) to aid in survival and progression. Previous work has shown that β1-integrin inhibitory antibodies can enhance the growth-inhibitory and apoptotic responses of human breast cancer cell lines to ionizing radiation, either when cells are cultured in three-dimensional laminin-rich ECM (3D lrECM) or grown as xenografts in mice. Here, we show that a specific α heter… Show more

“…Many studies indicate that integrin ␣5␤1-mediated adhesion is an advantage to tumor cells for survival, proliferation, migration, and even chemoresistance (20 -23, 46 -49). Because integrin ␣5␤1 expression positively correlates with tumor progression, it may be a promising therapeutic target for brain, breast, lung, and ovarian tumors (22)(23)(24)(25)(26). Activation of integrin ␣5␤1 by TNIIIA2 may cause unfavorable effects in the host.…”

“…Notably these variants are not found in soluble plasma fibrinogen, suggesting that these forms are more likely to polymerize [74]. Malignant cells express much higher levels of ED-A fibronectin and its receptor, α5β1 integrin, both of which have been linked to radiation resistance [75]. Likewise, tenascin-c also has multiple alternative splice forms, which are not observed in normal adult breast tissue [76].…”

“…Furthermore, the abberant ECM in tumors can alter fibronectin splicing even in non-malignant cells, whereas normalization of cell-matrix interactions in malignant cells normalizes fibronectin splicing [75]. Increased tissue stiffness appears to induce global changes in splicing, driving increased expression of ED-B fibronectin [78].…”

“…4 In tumor cells, integrins impact on proliferation/survival, therapy resistances and tumor recurrence. α5β1 integrin has emerged recently and has been shown to participate in aggressiveness of solid tumors [5][6][7][8][9] as reviewed in Schaffner et al 10 High expression of α5 integrin is associated with a decrease in patient overall survival, making α5β1 integrin an important factor in resistance to therapies. Numerous preclinical data support the ability of integrin antagonists to disrupt integrin signaling pathways leading to inhibition of angiogenesis and/or tumor growth but also to sensitization toward therapies.…”
