Oxidative stress plays a key role in breast cancer progression. However, little is still known about the relationship between the BRCA1 mutation, the incidence of breast cancer and oral homeostasis. This is the first study to evaluate the secretory function of salivary glands, biomarkers of redox balance, and oxidative damage to proteins and lipids in the saliva of subjects with the BRCA1 mutation. Ninety eight women were enrolled in the study and allocated to four groups based on molecular DNA testing: generally healthy patients without the BRCA1 mutation, patients with breast cancer but without the BRCA1 mutation, generally healthy patients with the BRCA1 mutation, and patients with both breast cancer and the BRCA1 mutation. We demonstrated that saliva from breast cancer patients with the BRCA1 mutation is characterized by enhanced antioxidant capacity and a higher degree of oxidative damage to proteins and lipids. The BRCA1 mutation can cause a predisposition to early salivary gland dysfunction, both in patients with breast cancer and in healthy individuals, leading to a decrease in salivary proteins. Using cluster analysis, we showed that salivary peroxidase, advanced glycation end-products (AGE), total antioxidant status (TAS) and malondialdehyde (MDA) may have particular clinical significance in non-invasive diagnostics of breast cancer.

The oral cancer constitutes 48% of head and neck cancer cases. Ninety percent of oral cancer cases are histologically diagnosed as oral squamous cell carcinomas (OSCCs). Despite new management strategies, the 5-year survival rate of oral cancer is still below 50% in most countries. Head and neck cancers are heterogeneous tumors, and this characteristic of them provides a challenge to treatment plan. Due to the poor outcomes in oral cancer, prevention is a necessity. In this review, a relevant English Literature search in PubMed, ScienceDirect, and Google Scholar from 2000 to mid-2018 was performed. All published articles related to oral cancer and its prevention were included. The risk factors of oral cancer and strategies of oral cancer prevention will be discussed.

Background: Oral cancer is one of the most prevalent malignancies and presents increasing incidence and mortality trends in the world, with a mortality rate of 14.33%. Oral squamous cell carcinoma (OSCC) constitutes 90% of all malignancies in the oral cavity. In spite of new therapeutic strategies, the risk of metastasis and mortality rate is still very high. Methods: A total of 54 samples (18 samples of each grade) were collected from the archives of the Pathology Department of Be'sat Educational Hospital, Hamadan, Iran from 2000 to 2017. The specimens were processed for immunohistochemistry (IHC) analysis. Besides, the samples were stained with Periodic Acid-Schiff (PAS) method for vasculogenic mimicry (VM) detection. Results: The chi-square test showed statistically significant differences between the expression level of endocan and lesion grade in the research groups (P = 0.025), lesion grade and VM formation (P = 0.001), and tumor grade and microvessel density (MVD) count (P = 0.000). Conclusions: The results suggest that endocan mediates tumor progression possibly via a few mechanisms such as cancer stem cells (CSCs), epithelial-mesenchymal transition (EMT), and VM formation. Further studies are required to reveal the mechanisms, by which the endocan acts as an oncogene. In addition, endocan could be considered as one of the tumor markers and a possible new target for cancer therapy.
