“…These results are in agreement with data from the literature showing that the 5-HT 2C antagonist property, shared by a number of other clinically active antidepressant agents of different classes (Bakish et al, 1993;Jenck et al, 1994;Ni and Miledi, 1997;Stefanski and Goldberg, 1997;Millan et al, 2000), cannot by itself support their full antidepressant-like activity.…”

“…This hypothesis is consistent with the suggestion that 5-HT 2C receptor blockers might exert antidepressant activity [27,29,[34][35][36][37]. In this respect, it is interesting to note that several antidepressant drugs have been shown to bind with submicromolar affinity to 5-HT 2C receptors in the pig brain and to antagonize mCPP-induced penile erections in rats, an effect mediated through the stimulation of central 5-HT 2C receptors [37][38][39]. Based on those findings, Matteo et al [34] have carried out experiments showing that acute administration of amitriptyline and mianserin, two antidepressants with high affinity for 5-HT 2C receptors, enhances DA release in the rat nucleus accumbens by blocking these receptor subtypes, in addition to their other pharmacological properties.…”

“…This hypothesis is consistent with the evidence that some antidepressant drugs display prominent 5-HT 2C receptor binding, e.g. tricyclics (amitriptyline, imipramine), mianserin, nefazodone, opipramole, trazodone [67,152,153] and fluoxetine [148,154]. Based on those findings, Di Matteo and colleagues [155] have carried out experiments showing that acute administration of amitriptyline and mianserin enhances DA release in the rat nucleus accumbens by blocking 5-HT 2C receptor subtypes.…”
