“…However, several additional publications suggest that bosentan may provide an effective and swift response in the treatment of digital ulcers [1319], as observed in the case herein presented. Macitentan also provided rapid (less than 3 months) and complete healing while maintaining a normal hepatic function and without observed DU recurrence after 6 months, arguing in favor of its use.…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…Bosentan is licensed for prevention rather than therapy of DUs, based on two randomized, double-blind, placebocontrolled trials; RAPIDS-1 and RAPIDS-2 (5,6). However, in some patients (7)(8)(9), in a retrospective study ( 10) and three prospective, open-label studies (2,11,12), it also had a curative effect. These clinical observations confirm the hypothesized role of endothelin in the pathogenesis of DUs.…”

“…Bosentan was able to reduce the development of new ulcers in the treatment versus the placebo group by 48% (1.4 vs. 2.7 new ulcers). Although no healing of preexistent ulcers has been observed in the RAPIDS-1 study, several authors reported on cases with a marked improvement of digital ulcers [3,4,5]. In a long-term prospective noncontrolled study, bosentan showed a significant decrease in the number of ulcers [6].…”
