Biological aspects of the DM28C clone of Trypanosoma cruzi after metacylogenesis in chemically defined media

Abstract: The biological characterization of the Trypanosoma cruzi clone Dm 28c in terms of its growth in LIT medium, cell-cycle, infectivity to mice and interaction with professional and non-professional phagocytic cells shows that it behaves as a bona fide T. cruzi representant. The biological properties of this myotropic clone do not change according to the origin of the trypomastigote forms (i. e., from triatomines, infected mice, cell-culture or from the chemically defined TAUP and TAU3AAG media). In addition Dm 28… Show more

“…Parasites stocks -The Tulahuen (Pizzi 1957), SA strains, Dm 28c clone (Contreras et al 1988) of T. cruzi and Choachí-2V strain of T. rangeli were used. The SA strain was isolated in 1999, from Rhodnius pallescenses, in the San Antonio region, Magdalena, Colombia, where an outbreak of acute Chagas disease occurred that year, with several fatal human cases.…”

“…T. cruzi clone Dm28c, isolated from opossum (Contreras et al, 1988) was used in this study. Parasites were maintained by weekly passages in liver infusion tryptose medium (LIT) (Camargo, 1964); epimastigotes were transformed into metacyclic trypomastigotes in artificial triatomine urine medium (TAU) (I90 mM-NaC1, 17 mM-KCl, 2 mM-MgCl,, 2 mM-CaC12, 8 mM-potassium phosphate buffer, pH 6.0) supplemented with 10 mM-L-proline (TAUP) as previously described (Contreras et al, 1985).…”

“…1985 ; Bonaldo et al, 1988). In addition to morphological changes, the metacyclogenesis process involves differential gene expression (Contreras et al, 1985;Bonaldo et al, 1988) and changes in fatty acid composition (Esteves et al, 1989).…”

“…Parasite culture and stress conditions -Epimastigotes of T. cruzi clone Dm28c (Contreras et al 1988) were maintained in the exponential growth phase by subculturing approximately 2 x 10 7 cells/mL every three days at 28ºC in liver infusion tryptose (LIT) medium supplemented with 10% foetal bovine serum (Camargo 1964). Epimastigotes in late growth phase were cultured for five days under the same conditions.…”
