The effect of gold nanoparticles (GNPs) synthesized in marine algae has been described in the context of skin, where they have shown potential benefit. Ecklonia stolonifera (ES) is a brown algae that belongs to the Laminariaceae family, and is widely used as a component of food and medicine due to its biological activities. However, the role of GNPs underlying cellular senescence in the protection of Ecklonia stolonifera gold nanoparticles (ES-GNPs) against UVA irradiation is less well known. Here, we investigate the antisenescence effect of ES-GNPs and the underlying mechanism in UVA-irradiated human dermal fibroblasts (HDFs). The DPPH and ABTS radical scavenging activity of ES extracts was analyzed. These analyses showed that ES extract has potent antioxidant properties. The facile and optimum synthesis of ES-GNPs was established using UV-vis spectra. The surface morphology and crystallinity of ES-GNPs were demonstrated using high resolution transmission electron microscopy (HR-TEM), energy dispersive spectroscopy (EDS), X-ray diffraction (XRD), and Fourier-transform infrared spectroscopy (FT-IR). ES-GNPs presented excellent photocatalytic activity, as shown by the photo-degradation of methylene blue and rhodamine B. A cellular senescence model was established by irradiating HDFs with UVA. UVA-irradiated HDFs exhibited increased expression of senescence-associated β-galactosidase (SA-β-galactosidase). However, pretreatment with ES-GNPs resulted in reduced SA-β-galactosidase activity in UVA-irradiated HDFs. Intracellular ROS levels and G1 arrest in UVA-irradiated HDFs were checked against the background of ES-GNP treatment to investigate the antisenescence effects of ES-GNPs. The results showed that ES-GNPs significantly inhibit UVA-induced ROS levels and G1 arrest. Importantly, ES-GNPs significantly downregulated the transcription and translation of MMP (matrix metalloproteinases)-1/-3, which regulate cellular senescence in UVA-irradiated HDFs. These findings indicate that our optimal ES-GNPs exerted an antisenescence effect on UVA-irradiated HDFs by inhibiting MMP-1/-3 expression. Collectively, we posit that ES-GNPs may potentially be used to treat photoaging of the skin.

Enriched mechanical, thermal, and blood compatibility of single stage electrospun polyurethane nickel oxide nanocomposite for cardiac tissue engineering

Electrospinning is a versatile technique for producing nanocomposite in biomedical applications. In this study, polyurethane (PU) cardiac patch loaded with nickel oxide (NiO) was fabricated using electrospinning technique. The morphology study revealed that the PU/NiO nanocomposites exhibited reduced fiber diameter (758 AE 157.10 nm) and pore diameter (868 AE 73.26 nm) compared to the pristine PU (fiber diameter 890 AE 116.91 nm and pore diameter 1064 AE 74.31 nm). The contact angle study indicated the decreased wettability behavior of the electrospun nanocomposite (106 AE 0.58 ) than the pure PU (100 AE 0.58 ). The incorporation of NiO into PU improved the mechanical strength (15.25 MPa) and surface roughness (448 nm) of the pristine PU (tensile strength 7.12 MPa and surface roughness 313 nm). The developed PU/NiO nanocomposites showed delayed blood clotting time and low hemolytic percentage as revealed in the coagulation studies insinuating the improved anticoagulant nature compared to the pristine PU. Moreover, 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt (MTS) assay demonstrated the nontoxic behavior of fibroblast cells in the developed nanocomposite (135 AE 1%) than the pristine PU (133.33 AE 8.96%). The newly developed nanocomposite patch rendered better physicochemical, improved blood compatibility, and nontoxic to the fibroblast cells. Hence, the electrospun PU/NiO nanocomposite might serve as a plausible scaffold for the cardiac tissue engineering.

This study investigates (i) in planta uptake and transfer of magnetic nanoparticles (MNPs) in the plant body and (ii) impact of MNPs on plant nutrition. For these purposes, barley (Hordeum vulgare L.) seedlings were subjected by varied MNP doses (125 to 1000 mg L −1 of CoNd 0.2 Fe 1.8 O 4) for 3 weeks in a hydroponic system. Plant tissues (root and leaf) were analyzed by using vibrating sample magnetometer (VSM) and inductively coupled plasma optical emission spectrometer (ICP-OES) techniques to understand MNPs' uptake and translocation in the plant body, and plant nutrition status as well. Elemental composition and magnetic behavior analyses of plant parts proved that MNPs, sized in 8.4 ± 0.05 nm, are uptaken by the plant roots and led to an increase in iron (Fe), neodymium (Nd), and cobalt (Co) contents of leaves (p < 0.005). However, compared with the untreated control, the amount of some macro-and micro-elements (K, Ca, Mg, Mn, and P) are declined in the leaf by increased MNP doses (p < 0.05). Root-to-leaf translocation index (%) of the elements were dramatically decreased, except the one for Fe which increased from 25 (control) to 55% in 1000 mg L −1 condition. Accordingly, MNPs are uptaken by the plant roots and transferred to the leaves. However, it suppresses the translocation of essential nutrients. This finding shows that MNPs used in this study is detrimental for plant mineral nutrition. Besides, the VSM technique coupled with ICP-OES enables to track MNPs in the plant body.
