Biofunctionalization of Fluorescent Rare‐Earth‐Doped Lanthanum Phosphate Colloidal Nanoparticles

Abstract: Approximately eight avidin molecules are conjugated to each Ce/Tb‐doped lanthanum phosphate nanoparticle (NP) through the formation of amide bonds (see scheme). This biofunctionalization process comprises modification of the LaPO4 NPs with 6‐aminohexanoic acid (AHA) to confer colloidal stability, activation of the AHA carboxy groups by 1‐ethyl‐3‐[3‐dimethylaminopropyl]carbodiimide, and avidin conjugation.

“…Again, the attachment of larger biomolecules can be observed by the increase of the total particle size as discussed before. In combination with organic fluorophores conjugated to the molecule of interest, the presence of those molecules on the particle surface can be accessed by the absorption or fluorescence emission (Demers et al 2000;Meiser et al 2004;Sari et al 2004;Hurst et al 2006), or by stepwise photobleaching of the organic dye (Casanova et al 2007). …”

“…A later work reported by the same group indicates that a luminescence quantum yield up to 61% can be achieved in this luminescent Ce 3+ /Tb 3+ codoped downconversion nanoparticles (Meyssamy et al, 2001)]. The reported low toxicity and luminescence stability allow these Ln 3+ doped LaPO 4 to be potentially used as biolabels towards clinical practices (Hutchison 1999;de Sousa Filho and Serra, 2011;Meiser et al, 2004 (Luwang et al, 2011;Mai et al, 2007;Yang et al, 2009;Li et al, 2009 (Balakrishnaiah et al, 2010). Both results suggest that the LnPO 4 material is promising host lattice for Ln 3+ ions to produce superior upconversion luminescence along the well-established fact on producing efficient downconversion luminescence.…”

“…Orthophosphate materials have been used for ceramic materials, catalysts, fluorescent materials, dielectric substances, metal surface treatment, detergent, food additives, fuel cells, pigments, etc. (Shuetz & Caruso, 2002;Meiser et al, 2004). The condensed phosphates have different properties from those of orthophosphates and can therefore be used as novel functional materials , Onoda et al, 2005Briche et al, 2010;Onoda et al, 2010).…”
