BackgroundEstrogen deficiency is an etiological factor of postmenopausal osteoporosis (PMO), which not only decreases bone density in vertebrae and long bone, but also aggravates inflammatory bone loss in alveolar bone. Recent evidence has suggested the critical role of gut microbiota in osteoimmunology, and modulation of gut microbiota may have positive influence on bone metabolisms. The present study aimed to evaluate the therapeutic effects of probiotics on alveolar bone loss under estrogen-deficient condition. Inflammatory alveolar bone loss induced by either chronic periodontitis or apical periodontitis was established in ovariectomized (OVX) rats, which were gavage-fed with probiotics daily until sacrifice. Gut microbiota and gut permeability, as well as alveolar bone loss and the related osteoimmune were evaluated to investigate the effects and underlying mechanisms by which probiotics counter the alveolar bone loss under estrogen-deficiency. ResultsWe found that administration of probiotics significantly prevented periodontal and apical bone resorption in OVX rats. Administration of probiotics significantly enriched butyrate-producing genera and enhanced gut butyrate production, resulting in improved intestinal barrier and decreased gut permeability in the OVX rats. Furthermore, the estrogen deprivation-induced inflammatory responses were suppressed in probiotics-treated OVX rats, as reflected by reduced serum levels of inflammatory cytokines and a balanced distribution of CD4+IL-17A+Th17 cells and CD4+CD25+Foxp3+Treg cells in the bone marrow. ConclusionOur data demonstrate that probiotics can effectively attenuate alveolar bone loss by modulating gut microbiota and further regulating osteoimmune, and thus represent a promising adjuvant in the treatment of alveolar bone loss under estrogen-deficiency.

Background Adequate sunlight exposure helps reduce bone loss and is important to bone health. Currently, about 90% of the world population spends a major portion of daily life under artificial lighting. Unlike sunlight, LED white light, the main source of artificial lighting, has no infrared radiation, which is known to be beneficial to human health. In artificial lighting environments, infrared supplementation may be used to simulate the effects of sunlight on bone metabolism. Results Here, we supplemented white LED exposure with infrared light in normal and ovariectomized rats for three consecutive months and examined bone turnover, bone mass, and bone density. We also analyzed the structure and function of gut microbiota in the rats. Infrared supplementation significantly reduced the abundance of Saccharibacteria and increased the abundance of Clostridiaceae 1 and Erysipelotrichaceae bacteria. Our results indicate that changes in the gut microbiome correlate well with bone mass and bone metabolism.Conclusions Our work demonstrates that infrared supplementation can have a positive effect on rat bone metabolism by affecting gut microbiota. Our findings will be important considerations in the future design of healthy lighting environments that prevent or possibly ameliorate osteoporosis.

Background Obesity has a negative impact on bone health; thus far, no specific pharmacotherapy has been approved. Berberine has shown improvements in osteoporosis in some animal models. However, it remains unknown whether berberine ameliorates obesity-induced bone damage. This study aims to evaluate the effect of berberine on bone damage in high-fat diet (HFD)-induced obese rats. Methods Male Sprague-Dawley rats were randomly assigned to a normal control diet (ND) group or HFD group. After the HFD induced obesity, the models were successfully established. Then, these rats were randomly divided into the HFD + berberine (HB) group or HFD group and were intraperitoneally administered berberine or an equivalent volume of DMSO, respectively, once a day for another 10 weeks. Micro-CT and three-point bending tests were conducted to evaluate bone microstructure and biomechanics. Serum was collected for the detection of P1NP, CTX-1, calcium (Ca) and phosphorus (P), tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β). HE staining was performed to observe the number of trabecular and bone marrow adipocytes. Results HFD-induced obese rats displayed decreased biomechanical properties, such as maximum load, maximum fracture load, ultimate tensile strength, stiffness, and energy absorption, compared with the ND rats (p<0.05). However, berberine attenuated bone damage, including maximum load, maximum fracture load, and stiffness, in the HB group compared with the HFD group (p<0.05). Trabecular bone markers were decreased in the HFD group compared with the ND group (p<0.05). All parameters were improved, as shown by micro-CT and HE staining, in the HB group compared to the HFD group. Cortical bone markers were not significantly different among all groups. Moreover, CTX-1, TNF-α, IL-1β and the number of adipocytes in bone marrow were significantly increased in HFD-induced obese rats. After treatment with berberine, TNF-α, IL-1β and the number of adipocytes in bone marrow were significantly decreased, and P1NP levels were higher in the HB group than in the HFD group. Conclusions Berberine might be a potential therapeutic agent for treating bone damage in HFD-induced obese rats by inhibiting inflammatory factors, reducing bone marrow adiposity and improving bone formation.
