Bafilomycin A1 Treatment Retards Transferrin Receptor Recycling More than Bulk Membrane Recycling

Abstract: After internalization via clathrin-coated pits, endocytosed material rapidly enters sorting endosomes, organelles that sort recycled molecules such as transferrin (Tf) 1 bound to the transferrin receptor (Tf-R) from lysosomally destined molecules such as low density lipoprotein (1-3). We have provided evidence for a sorting mechanism in which components not carrying special retention or targeting signals are removed from the sorting endosome by a default process based on the geometry of the sorting endosome an… Show more

“…1) suggest that alkalinization of intramembrane space may not acutely block translocation of the preexisting IRVs but may interfere with the formation of this compartment. Such a hypothesis is consistent with several earlier reports showing that bafilomycin A1 inhibits budding of small vesicles from donor endosomal membranes (1,5,25).…”

“…To further examine the role of recycling in maintaining DAT surface levels, we blocked constitutive recycling with the vacuolar ATPase inhibitor bafilomycin A 1 , which blocks TfR recycling and reduces TfR surface expression (26). Bafilomycin treatment (0.5 M) for 30 min reduced TfR plasma membrane expression to 60.6 Ϯ 6.6% of that observed for vehicle-treated cells (Fig.…”

“…First, azithromycin delayed sequestration in clathrin-coated pits of receptor-bound ligands, as best evidenced for PAP/Fc␥ receptor complexes (ϳ10 min), but did not appreciably affect the subsequent rate of endocytosis per se. Second, azithromycin causes a major delay of transferrin-receptor recycling (ϳ10 min) without impairing the subsequent rate of efflux, which contrasts with the absence of reported delay but prolonged half-life of efflux of transferrin receptors in cells treated with monensin and bafilomycin A1 [7,23,24], two drugs known to markedly impair acidification of endosomes and lysosomes [71,72].…”
