Automated genomic context analysis and experimental validation platform for discovery of prokaryote transcriptional regulator functions

Abstract: BackgroundThe clustering of genes in a pathway and the co-location of functionally related genes is widely recognized in prokaryotes. We used these characteristics to predict the metabolic involvement for a Transcriptional Regulator (TR) of unknown function, identified and confirmed its biological activity.ResultsA software tool that identifies the genes encoded within a defined genomic neighborhood for the subject TR and its homologs was developed. The output lists of genes in the genetic neighborhoods, their… Show more

“…To further explore the role of Ms6243, we carried out an extensive search of the literature and found many homologs of Ms6243 in different organism, adapting various functions. One of them we found is Bxe_B3017 from Burkholderia xenovorans , the function of which is unknown, that shares 78% sequence identity with Ms6243 [31]. bxe_B3017 is the neighbouring gene of bxe_B3018 , a TetR family transcriptional regulator that shares its 39% of amino acid sequence with that of Ms6244.…”
