Autism and Möbius sequence: an exploratory study of children in northeastern Brazil

Abstract: -The psychiatric examination was performed with diagnostic instruments for autism (DSM-IV and Childhood Autism Rating Scale-CARS) in 23 children with Möbius sequence. From the 23 patients studied with Möbius sequence, five (26.1%) met the diagnostic criteria for infantile autism according DSM-IV and two (8.6%), under two years old, showed autistic-like behavior. The scores for six children were compatible to severe autism symptoms according CARS and one child met the criteria for moderate autism symptoms. Amon… Show more

“…As noted previously, autism risk factors include environmental challenges, such as maternal use of pharmaceutical agents with neurotoxic effects, [226][227][228]241,242 prenatal exposure to viral infections or maternal stress 233,234 and, in addition, exposure to high levels of environmental pollutants, including heavy metals. 275,276 A recent review by Herbert et al 7 identified 135 genes that have been shown to mediate responses to environmental challenge, and that are located within autism linkage regions.…”

“…235,238,239 Exposure to teratogens during gestation has also been shown to be a risk factor for autism, with higher disease incidence associated with maternal use of valproic acid, [226][227][228]240 thalidomide 241 and misoprostol. 242 The detrimental neurodevelopmental effects of maternal infection have been attributed, in part, to the induction of inflammatory cytokines. 243 Mouse models for prenatal exposure to maternal infection or inflammation have shown that the challenged offspring demonstrate an altered behavioral phenotype, including deficiencies in social interaction, exploration and sensorimotor gating.…”

“…In most studies, Moebius sequence has been defined as a congenital facial weakness with congenital impairment of ocular abduction [1,7,27,28].…”

“…So far, four studies with 17-37 Moebius patients [1,11,12,27] showed a markedly increased incidence of autism spectrum disorder (ASD) (5-29 vs. 0.63% in the normal population [8]). Nevertheless, the association of Moebius sequence with ASDs seems to be questionable as some of these studies showed methodological problems, and mental retardation was overrepresented in at least two of them [6].…”

“…In addition, in more than 50% of cases, the environmental agent that caused a fetal anomaly is not identified (3) . Misoprostol, alcohol, cigarettes, marijuana, cocaine, and rubella have been associated with Möbius sequence (7,10,13,(16)(17)(18)20,21,29,30) . In the present study, teratogens could be identified in most cases (56%), and 9.8% of the mothers were exposed to more than one teratogen.…”
