Abnormal distributions of transition metals inside the brain are potential diagnostic markers for several central nervous system diseases, including Alzheimer's disease (AD), Parkinson's disease, dementia with Lewy bodies (DLB), bipolar disorders and depression. To further explore this possibility, the total concentrations of iron, zinc, copper, manganese, aluminum, chromium and cadmium were measured in post-mortem hippocampus and amygdala tissues taken from AD, DLB and Control patients. A statistically significant near fifty percent reduction in the total copper levels of AD patients was observed in both the hippocampus and amygdala. The statistical power of the hippocampus and amygdala copper analysis was found to be 86 and 74% respectively. No statistically significant deviations in the total metal concentrations were found for zinc, manganese, chromium or aluminum. Iron was found to be increased by 38% in AD amygdala tissues, but was unchanged in AD hippocampus tissues. Accounting for differences in tissue water content, as a function of both tissue type and disease state, revealed more consistencies with previous literature. To aid in the design of future experiments, the effect sizes for all tissue types and metals studied are also presented.

This work considers the development of a novel analytical procedure for rapid and simple size fractionation of metal species from human serum samples, with a great potential for studying characteristic abnormalities of metal homeostasis associated with Alzheimer's disease and other neurodegenerative disorders. For this purpose, serum samples are subjected to protein precipitation under non-denaturing conditions, taking special care for maintaining the integrity of metal-biomolecule bindings and for preventing species transformation, and then both the supernatant (low molecular mass fraction) and the precipitate (high molecular mass fraction) are analyzed by ICP-MS in order to determine the biodistribution of serum trace elements. This methodology is validated for 11 trace elements, including aluminum, cadmium, cobalt, chromium, copper, iron, manganese, molybdenum, selenium, vanadium, and zinc, in terms of sensitivity, selectivity, accuracy, and precision.

Introducción: Los elementos traza u oligoelementos son micronutrientes disponibles en mínima cantidad en el cuerpo humano y constituyen funciones vitales, lo que les confiere su importancia. El exceso o déficit en las concentraciones de oligoelementos como zinc, litio, cobre y plomo podrían estar involucrados en las diferentes fases del trastorno afectivo bipolar. Objetivo: Analizar la información actual disponible en la literatura sobre la relación entre elementos traza y el trastorno afectivo bipolar. Métodos: Búsqueda en paralelo por dos revisores en las bases de datos Ovid, Pubmed, SciELO y Cochrane. Resultados: Se encontraron un total de 47 artículos publicados entre 1988 y 2015, aunque al final se evaluaron 12 artículos en texto completo. Conclusiones: La literatura disponible sugiere asociación entre altas cantidades de zinc en fase maniaca y escasa presencia en la fase depresiva de pacientes con trastorno afectivo bipolar. No se encontró asociación significativa con los otros oligoelementos.
