Assessment of DNA damage using chromosomal aberrations assay in lymphocytes of waterpipe smokers

Abstract: Objectives The aim of this study was to investigate the genotoxicity of waterpipe smoking in the lymphocytes of waterpipe smokers using chromosomal aberrations (CAs) assay. Materials and Methods Fifty waterpipe smokers and 18 healthy non-smokers volunteered to participate in the study. Additionally, 18 heavy cigarette smokers were recruited for comparison. Chromosomal aberrations (CAs) assay was used to evaluate DNA damage in the lymphocytes. Results The results showed that similarly to cigarette smoking, … Show more

“…On the other hand, previously reported higher benzene and carcinogenic PAHs from water pipe smoking suggest that the health risks associated with these toxicants are likely higher among water pipe smokers than nonsmokers with secondhand cigarette smoke exposure or even among light and intermittent cigarette smokers. In vitro studies show that water pipe smoke causes DNA damage, has cytotoxic and mutagenic effects, and causes endothelial dysfunction (49-51). Water pipe smoking compromised cardiac autonomic function in a clinical study (52).…”

“…For example, nicotine has been found to improve performance on attention and memory tasks (Rezvani and Levin, 2001), and to attenuate memory impairment associated with several health conditions in both human and animals (Harris et al, 2004; White and Levin, 2004; Aleisa et al, 2006; Carrasco et al, 2006; Jubelt et al, 2008; Aleisa et al, 2011; Srivareerat et al, 2011; Alzoubi et al, 2013c). In addition to nicotine, the high levels of carbon monoxide inhaled during WTS in human, sometimes enough to cause acute toxicity (Eissenberg, 2013; von Rappard et al, 2014), and induce oxidative stress in both human and animals (Valenca et al, 2008; Khabour et al, 2011; Alsatari et al, 2012; Khabour et al, 2012a), thus, probably influencing cognitive function.…”

“…For example, waterpipe smoking has been shown to be associated with increases in the frequencies of sister chromatid exchanges and chromosomal aberrations in lymphocytes obtained from smokers (Yadav and Thakur, 2000;Khabour et al, 2011;Alsatari et al, 2012). In addition, the level chromosomal damage was found to be strongly correlated with the magnitude of waterpipe use (Khabour et al, 2011;Alsatari et al, 2012). Moreover, waterpipe smoking has been reported to induce micronuclei in buccal mucosa cells (Boulos et al, 2009).…”

“…Recent studies found that acute exposure to waterpipe smoke caused lung inflammation and oxidative stress to same degree to that induced by cigarette smoking (Khabour et al, 2012a). Moreover, the genotoxicity associated with waterpipe use is significantly higher in heavy waterpipe users than in heavy cigarette smokers (Khabour et al, 2011;Alsatari et al, 2012).…”
