The investigation on antimicrobial mechanisms is a challenging and crucial issue in the fields of food or clinical microbiology, as it constitutes a prerequisite to the development of new antimicrobial processes or compounds, as well as to anticipate phenomenon of microbial resistance. Nowadays it is accepted that a cells population exposed to a stress can cause the appearance of different cell populations and in particular sub-lethally compromised cells which could be defined as viable but non-culturable (VBNC). Recent advances on flow cytometry (FCM) and especially on multi-parameter flow cytometry (MP-FCM) provide the opportunity to obtain high-speed information at real time on damage at single-cell level. This review gathers MP-FCM methodologies based on individual and simultaneous staining of microbial cells employed to investigate their physiological state following different physical and chemical antimicrobial treatments. Special attention will be paid to recent studies exploiting the possibility to corroborate MP-FCM results with additional techniques (plate counting, microscopy, spectroscopy, molecular biology techniques, membrane modeling) in order to elucidate the antimicrobial mechanism of action of a given antimicrobial treatment or compound. The combination of MP-FCM methodologies with these additional methods is namely a promising and increasingly used approach to give further insight in differences in microbial sub-population evolutions in response to antimicrobial treatments.

The realization of antimicrobial films through the incorporation of active agents into a polymer matrix is a promising alternative to the direct addition of antimicrobials into the food matrix. To this aim, the goal of this work was to develop a sustainable, food packaging solution with antimicrobial effectiveness and high functional performance, based on Ethyl-Nα-dodecanoyl-L-arginate (LAE). Active biodegradable coatings, easy soluble to be removed, were realized by spreading a Polylactic acid/LAE coating solution, at different antimicrobial concentration (from 0 to 20%), on a recyclable Polyethylene-terephthalate substrate. The antimicrobial activity of the multilayer films was tested in vitro against E.Coli CECT 434 strain as pathogenic agent in liquid culture media. Moreover, the LAE chemical interaction with the PLA matrix was investigated, as well as its effect on the adhesion, wetting, optical and barrier properties of the films. The results pointed out that that the minimum LAE concentration incorporated already guarantees an antimicrobial activity comparable to commercial antimicrobial packaging solutions, and that, among the systems investigated, 10% LAE is the minimum concentration guaranteeing total inhibition without significantly altering the functionalities of the developed systems.

BACKGROUND: Citrus 'Benimadonna' (Citrus reticulata × Citrus sinensis) is a high-value perishable fruit; thus there is an urgent need for a preservation technology with high effectiveness and low safety risk from industries. N⊍-Lauroyl-L-arginine ethyl ester hydrochloride (LAE) was applied to enhance preservability by compounding with natamycin, and a possible fungal inhibition mechanism based on the hypothesis of an impact on the cell membrane by surfactant was investigated. RESULTS:In vitro testing showed that the minimum inhibitory concentration of LAE against Penicillium expansum (PE), isolated as the predominant spoilage-inducing fungus, was 32 mg L −1 and it was partially synergistic with natamycin. Subsequent in vivo testing proved the inhibition capacity. During 90 days' refrigerated preservation, spoilage rate was significantly decreased by preharvest spraying versus control without extra taste loss, and LAE showed an alleviating benefit on total pectin loss. Subsequently, electron microscopic imaging and intracellular protein levels of PE exposed to LAE indicated that LAE stress led to increased permeability and decreased cell integrity. Moreover, peroxidase, superoxide dismutase and catalase revealed that LAE enhanced oxidative stress, while pectinase was antagonized.CONCLUSION: The present investigation first introduced LAE as a candidate active ingredient for citrus preservative. A theoretical basis was provided for the development of preservation technology for high-value perishable fruit. According to the authors' knowledge this study is the first report on the inhibition mechanism of LAE in terms of oxidative stress.
