ASCT2 silencing regulates mammalian target-of-rapamycin growth and survival signaling in human hepatoma cells

Abstract: System ASC amino acid transporter-2 (ASCT2) was previously demonstrated to be essential for human hepatoma cell growth and survival, as its silencing via inducible antisense RNA expression results in complete apoptosis within 48 h by a mechanism that transcends its role in amino acid delivery. To gain mechanistic insights into the reliance of cancerous liver cells on ASCT2, the aim of this study was to determine the early consequences of its silencing on the growth and survival signaling that presage apoptosis… Show more

“…AtCOP10 and AtUEV1B were amplified from Columbia-0 cDNA and HsMUB from SK-HEP cDNA (38) and similarly cloned into pENTR/D-TOPO. The AtCOP10 splice variant At3g13550.1 was gel-excised prior to D-TOPO cloning.…”

“…Slc1a5 is up-regulated in multiple types of cancer and has been implicated in mediating net glutamine uptake in cancer cells (Fuchs and Bode 2005). Glutamine uptake into the cell through Slc1a5 has been shown to be essential for regulating mTOR activity (Fuchs et al 2007;Nicklin et al 2009) by making it available for coupling glutamine export to leucine import into the cell through Slc7a5 (Nicklin et al 2009). Slc6a19 preferentially transports neutral amino acids, including leucine, isoleucine, and glutamine.…”
