Ascorbate synthesis and ascorbate peroxidase activity during the early stage of wheat germination

Abstract: O. 1997, Ascorbate synthesis and ascorbate peroxidase activity during the early stage of wheat germination. -Physiol. Plant. 100: 894-900.Embryos from dry caryopses of wheat {Triticum durum L. cv. Norba) are completely devoid of ascorbate (ASC) but contain a low amount of dehydroascorbate (DHA). The de novo biosynthesis of ASC starts in the wheat embryos after 8-10 h of germination but before the ASC biosynthetic pathway is completely restored the embryos can provide themselves with ASC by the reduction of the… Show more

“…The high activity of the ASC recycling enzyme MDHAR and the increasing levels of MDHA and DHA in BSO-treated embryos may be a strategy to eYciently regenerate ASC at the onset of germination, prior to the complete reactivation of the de novo ASC biosynthetic pathway. These metabolic alterations, documented during normal embryonic growth in vivo and in vitro (De Gara et al 1997;Stasolla and Yeung 2001), are not observed in control embryos. The potential to quickly regenerate ASC from MDHA in the presence of an oxidized glutathione environment may explain the improved conversion frequency of BSO-treated embryos (Belmonte et al 2006).…”

“…Soluble proteins were extracted from 2 g of a 3-d-old BY-2 culture with 20 mL of the buffer described in De Gara et al (1997). Native gel electrophoresis and subsequent staining for GSSG reductase activity was performed according to Foyer et al (1991).…”

“…It has been widely reported that APX plays a key role in removing the toxic H 2 O 2 in plant cells, 35,36,37 the correct temporal expression of APX has to be an important factor for seed germination. 38 APX activity in the two cultivar seeds disappeared at the end of their development and did not show any significant differences during seed maturation (Fig. 5A).…”
