Applications of Protein Microarrays in Biomarker Discovery for Autoimmune Diseases

Abstract: Dysregulated autoantibodies and cytokines were deemed to provide important cues for potential illnesses, such as various carcinomas and autoimmune diseases. Increasing biotechnological approaches have been applied to screen and identify the specific alterations of these biomolecules as distinctive biomarkers in diseases, especially autoimmune diseases. As a versatile and robust platform, protein microarray technology allows researchers to easily profile dysregulated autoantibodies and cytokines associated with… Show more

“…The membrane-based protein array, which is known to be a powerful platform for simultaneously detecting the expression levels of 507 human target proteins, has been proposed as a clinically useful method to identify disease biomarkers for diagnosis, prognosis, prediction, and understanding of previously unknown mechanisms involved in certain diseases [9][10][11]. However, to date, no study has used protein-antibody array technology to identify biomarkers for pregnancy outcome after placement of rescue cerclage for acute cervical insufficiency.…”

“…Peptide-based microarrays are similar to protein microarrays, but they have been constructed using thousands of pre-synthesized peptides on a microarray surface ( Li et al, 2021 ). Compared to proteins, synthesized peptides are more stable, easy to produce and modify, and more importantly, less expensive.…”

“…The peptide-based microarray technique required a low amount of human serum sample and showed higher sensitivity compared to the conventional methods. Therefore, it could be adopted as a cost-effective screening method for the identification of other lentil allergen epitopes ( Kühne et al, 2015 , Li et al, 2021 ).…”

“…Protein interactions, such as protein binding and enzyme-substrate reactions, may be studied using functional protein microarrays [26]. Purified recombinant proteins or peptide fragments are assembled into functional arrays that may be utilized for discovery-based research, such as the investigation of protein-protein, protein-DNA, protein-lipid, protein-drug and protein-peptide interactions [27].…”

“…Its benefit is that it just takes a little number of proteins to simultaneously investigate numerous analytes inside the original sample. As a result, protein microarrays are extremely effective in the identification of biomarkers [27].…”
