Applications of iTRAQ and TMT Labeling Techniques to the Study of Neurodegenerative Diseases

Abstract: : Neurodegenerative diseases are caused by progressive lesions or loss of specific nerve cells, which endanger human health. However, the mechanism by which neurodegeneration manifests remains unclear. Proteomics can shed light on this question as well as help establish diagnostic standards and discover new drug targets. The power of proteomics for understanding neurodegenerative diseases has increased substantially with the application of iTRAQ and TMT labeling techniques. This review focuses on progress in … Show more

“…Proteins display a wide range of variation in abundance, making low-abundant proteins easily masked by high-abundant proteins with similar mass or chemical properties (Smith et al, 2019). Comparative proteomics methods based on mass spectrometry, such as Label-free, iTRAQ, SWATH, PRM, allow us to identify proteins with significantly changed expression levels under specific conditions on a large scale (Bourmaud et al, 2016;Li et al, 2020). Joint analysis of proteomics and other omics provide more useful information for exploring the mechanisms of wool bending and hair follicle development.…”

“…Together with the technological advancement on LC-MS and bioinformatics software, it is easier to identify qualified proteins in these days. Regarding the quantification, the Isobaric Tag for Relative and Absolute Quantitation (iTRAQ) has been widely used in mammalian studies [ 46 ]. On the other hand, the application in fishes is still very limited.…”

“…Given that there is a limited understanding of AMH-associated secondary hypertension ( 24 , 25 ), this study mainly investigated the proteome profiles of AMH as well as adjacent control tissues using iTRAQ quantitative analysis, an advanced method with maximum protein coverage and precise quantitation ( 26 28 ). It turned out that 782 kinds of differentially expressed proteins were identified in AMH tissue, including 357 types of upregulated proteins and 425 types of downregulated proteins.…”
