RESUMENObjetivos. Evaluar el efecto inmunomodulador sobre poblaciones linfocitarias, células dendríticas (DC), citoquinas Th1/Th2/Th17 (T-helper) e inflamatorias en el ámbito sistémico y/o en el microambiente tumoral de ratones con o sin melanoma. Materiales y métodos. Se obtuvieron muestras de sangre periférica y/o de tumores primarios de ratones con melanoma B16 tratados o no con un extracto hidroalcohólico de Uncaria tomentosa (UT) con 5,03% de alcaloides oxindólicos pentacíclicos (UT-POA) obtenido de la corteza de la planta. Todos los ensayos de medición de células y citoquinas fueron realizados por citometría de flujo. Resultados. UT-POA a nivel sistémico incrementa la relación CD4/CD8a (Cluster of Differenciation), mientras que la activación celular es inversamente proporcional; incrementa la proporción de DCm (DC mieloides); induce un perfil Th1 proinflamatorio y reduce la respuesta Th17. TNF-a (tumor necrosis factor alpha) y IL-17A (interleuquina) correlacionan positiva y negativamente con la relación CD4/CD8a. Conclusiones. El incremento de Th1 (TNF-a) puede tener como consecuencia el incremento de linfocitos CD4 o la activación de macrófagos M1. Aunque UT-POA muestra un incremento de DCm, este no es dosis-dependiente. La disminución de Th17 (IL-17A) puede favorecer el funcionamiento de los linfocitos CD8a. UT-POA muestra mejores efectos inmunomoduladores en el ámbito sistémico que intratumoral. LYMPHOCYTE SUBSETS, DENDRITIC CELLS AND CYTOKINE PROFILES IN MICE WITH MELANOMA TREATED WITH Uncaria tomentosa ABSTRACT Objectives.To evaluate the immunomodulatory effect on lymphocyte subsets, dendritic cells (DC), Th1 / Th2 / Th17 and inflammatory cytokines on systemic level and/or in the tumor microenvironment of mice with or without melanoma. Materials and methods: Peripheral blood and/or primary tumors samples were obtained of mice with B16 melanoma treated or not with a hydroalcoholic extract of Uncaria tomentosa (UT) with 5.03% of pentacyclic oxindole alkaloids (UT-POA) obtained from the bark of the plant. All cell assays and cytokine measurements were performed by flow cytometry. Results. UT-POA systemically increased CD4/CD8a relation while cell activation was inversely proportional; increased the proportion of DCm; induced a pro-inflammatory Th1 profile and reduced Th17 response. TNF-α and IL-17A positively and negatively correlated with CD4/CD8a relation. Conclusions. The increase of Th1 (TNF-α) may result in the increase of CD4 or M1 macrophage activation. Although UT-POA shows increased DCm, is not dose-dependent. Th17(IL-17A) decreased can support the function of CD8a lymphocytes. UT-POA shows better systemic immunomodulatory effects than intratumoral.

Simira gardneriana is a Rubiaceae species commonly found in the Brazilian Northeast region, presented several therapeutic and biotechnological applications. In this paper, the antioxidant and photoprotective properties of extracts from the seeds of S. gardneriana were highlighted. The antioxidant activity of ethanol and methanol extracts (Si-EtOH and Si-MeOH, respectively) was determined, using 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) free radical scavenging assay. The photoprotective activity of the extracts was evaluated using a spectrophotometric method. Total phenolic and flavonoid content was determined. In addition, a qualitative analysis of phytochemical markers and a high-performance liquid chromatography-diode-array detector (HPLC-DAD) analysis were also performed for both extracts. Concerning the antioxidant activity in vitro, Si-MeOH (EC 50 = 70.94±2.17 µg/ml) showed greater activity in comparison to Si-EtOH (EC 50 = 138.60±7.39 µg/ml). Once the sun protection factor spectrophotometric (SPF spectrophotometric ) of samples was calculated, it was demonstrated that the extracts show a similar photoprotective effect on all concentrations tested. SiEtOH and Si-MeOH tested on a concentration of 100 mg/l, exhibited SPF values equal to 3.37±0.006 and 3.36±0.007, respectively. HPLC fingerprints was recorded and showed the presence of characteristic peaks for phenolic compounds. The extracts showed significant phenols and flavonoids content according to the quantification methods used. According to the results, it was concluded that Si-EtOH and Si-MeOH have significant antioxidant and photoprotective activities. These activities are probably related to the profile of flavonoids and phenolic compounds found in this species.

Purpose:The aim of this research was to develop and optimize a process for obtaining poly ε-caprolactone (PCL) nanoparticles loaded with Uncaria tomentosa (UT) extract. Methods: Nanoparticles were produced by the oil-in-water emulsion solvent evaporation method. Preliminary experiments determined the initial conditions of the organic phase (OP) and of the aqueous phase (AP) that would be utilized for this study. Ultimately, a three-factor three-level Box-Behnken design (BBD) was employed during the optimization process. PCL and polyvinyl alcohol (PVA) concentrations (X 1 and X 2 , respectively) and the AP/OP volume ratio (X 3 ) were the independent variables studied, while entrapment efficiency (Y 1 ), particle mean diameter (Y 2 ), polydispersity (Y 3 ), and zeta potential (Y 4 ) served as the evaluated responses. Results: Preliminary experiments revealed that the optimal initial conditions for the preparation of nanoparticles were as follows: OP composed of 5 mL ethyl acetate/acetone (3/2) mixture containing UT extract and PCL, and an AP of buffered PVA (pH 7.5) solution. Statistical analysis of the BBD results indicated that all of the studied factors had significant effects on the responses Y 1 , Y 2 , and Y 4, and these effects are closely described or fitted by regression equations. Based on the obtained models and the selected desirability function, the nanoparticles were optimized to maximize Y 1 and minimize Y 2 . These optimal conditions were achieved using 3% (w/v) PCL, 1% (w/v) PVA, and an AP/OP ratio of 1.7, with predicted values of 89.1% for Y 1 and 280 nm for Y 2 . Another batch was produced under the same optimal conditions. The entrapment efficiency of this new batch was measured at 81.6% (Y 1 ) and the particles had a mean size of 247 nm (Y 2 ) and a polydispersity index of 0.062 (Y 3 ). Conclusion: This investigation obtained UT-loaded nanoparticle formulations with desired characteristics. The BBD approach was a useful tool for nanoparticle development and optimization, and thus should be useful especially in the realm of phytotherapeutics, in which varied compositions may be assessed in quantitative and qualitative terms.
