In this work, a comparison study on active sites (the total phenolic, total flavonoids, and total triterpenes contents) and antioxidant activities (DPPH, ABTS, and FRAP assays) of different fractions from Pyrus ussuriensis Maxim was evaluated. Moreover, the inhibition capability on human hepatocarcinoma cells Bel‐7402 cells and the mechanism was also discussed. Results showed that the ethyl acetate fraction significantly scavenged DPPH and ABTS+ radicals, exhibited ferric ion reducing antioxidant, and inhibited Bel‐7402 cells proliferation. In addition, oleanolic acid was the dominant compound act on the Bel‐7402 cells in the extract and it induced apoptosis by the caspase pathway and induced cell cycle arrest at the G0/G1 phase by inhibiting the cyclin D1/CDK4 pathway. The extracts of P. ussuriensis Maxim were confirmed to have anti‐oxidative and antiproliferative effects against Bel‐7402 cell in vitro. Practical applications Fruits and vegetables which contain high levels of antioxidants can help to reduce the risk of chronic diseases such as cancer. Pyrus ussuriensis Maxim is a kind of edible and medical fruit with multiple bioactivities, whereas the capability to anti‐lung cancer activity has not been investigated. The extracts of P. ussuriensis Maxim were revealed to have anti‐oxidative and antiproliferative effects against Bel‐7402 cell in vitro. Accordingly, it is the first time to verify that oleanolic acid was the main chemical components of P. ussuriensis with antitumor potential.

Phytosterols, found in many commonly consumed foods, exhibit a broad range of physiological activities including anti-inflammatory effects. In this study, the anti-inflammatory effects of ergosterol, β-sitosterol, stigmasterol, campesterol, and ergosterol acetate were investigated in lipopolysaccharide (LPS)-induced RAW264.7 macrophages. Results showed that all phytosterol compounds alleviated the inflammatory reaction in LPS-induced macrophage models; cell phagocytosis, nitric oxide (NO) production, release of tumor necrosis factor-α (TNF-α), and expression and activity of pro-inflammatory mediator cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), and phosphorylated extracellular signal-regulated protein kinase (p-ERK) were all inhibited. The anti-inflammatory activity of β-sitosterol was higher than stigmasterol and campesterol, which suggests that phytosterols without a double bond on C-22 and with ethyl on C-24 were more effective. However, inconsistent results were observed upon comparison of ergosterol and ergosterol acetate (hydroxy or ester group on C-3), which suggest that additional research is still needed to ascertain the contribution of structure to their anti-inflammatory effects.

BackgroundTo establish the combination of doxorubicin (DOX) and silybin (SLB) in oral hepatic-targeting liposomes with the goal of reducing cardiotoxic side effects and improve oral hepatoma treatment.MethodsDistearoylphosphatidylethanolamine–polyethylene glycol–cholic acid-modified liposomes (CA-LP) were used to encapsulate DOX and SLB (CA-LP–DOX/SLB), and the hepatic targeting, efficacy against hepatoma and cardioprotective effects were evaluated by cell toxicity, scratch and apoptosis in vitro studies, and pharmacokinetics and pharmacodynamics in vivo studies.ResultsIn vitro cell studies showed that CA-LP–DOX/SLB inhibited HepG2 cell proliferation and HCC97H cell migration, and protected H9c2 cells. In vivo pharmacokinetics demonstrated that the CA-LP–DOX/SLB-treated group showed higher liver accumulation and lower heart accumulation of DOX relative to those in the CA-LP–DOX and LP–DOX-treated groups. In vivo pharmacodynamic studies showed that the CA-LP–DOX/SLB-treated group not only efficiently inhibited growth but also induced significantly less tissue damage than that observed in the CA-LP–DOX-treated group.ConclusionConcurrent administration of DOX and SLB via CA-LP provided a viable strategy to mitigate acute DOX-induced cardiotoxicity.
