Zanthoxylum zanthoxyloides, syn. Fagara zanthoxyloides, is a tree growing in West Africa and is used in traditional medicine against a variety of diseases, including malaria. In the work reported here, root bark and stem bark extracts of this tree, as well as compounds isolated from the extracts, have been investigated for activity in vitro against chloroquine-sensitive and chloroquine-resistant strains of Plasmodium falciparum. In addition, toxicity against nauplii of the brine shrimp Artemia salina has been studied. Dichloromethane extracts of the root bark and stem bark, and a methanol extract of the stem bark, showed anti-parasitic activity towards chloroquine-sensitive as well as chloroquine-resistant P. falciparum, with IC50 values between 1 and 10 µg/mL. Among the isolated compounds, bis-dihydrochelerythrinyl ether, buesgenine, chelerythrine, γ-fagarine, skimmianine, and pellitorine were the most active, with IC50 values of less than 5 µg/mL. The dichloromethane extracts were toxic to brine shrimp nauplii, with LC50 values of less than 1 µg/mL. Methanol extracts were much less toxic (LC50 between 50 and 100 µg/mL). Among the isolated substances, bis-dihydrochelethrinyl ether was the most toxic (LC50 ca. 2 µg/mL).

In this present study, in vivo antiplasmodial and antioxidant activities of Acacia gourmaensis A. RICH. (Mimosaceae) were evaluated based on its total extracts (ethanol-water, decoction, acetone), fractions (hexane fraction, ethyl acetate fraction, butanol fraction and aqueous fraction) and mineral contents. Total extract of decoction, acetone and EtOH-H2O fractions were used to evaluate chemical composition using HPLC-GC/MS-DAD methods. This plant is used in traditional medicine against liver disease, fractures, coughing fits and especially paludism. The antioxidant property was analyzed using 3 methods such as total antioxidant activity (TAC), iron reducing power (IRP) and radical scavenging activity (DPPH*). Antiplasmodial activity was performed in mice using red blood infected by Plasmodium berghei. The biological activities (antioxidants and antiplasmodial) were directed against plant phytochemical content 9 polyphenolic compounds were highlighted in the HPLC-DAD method, against 3 in HPLC-GC/MS. EtOH-H2O gave the best anti-TAC with 6.53 ± 0.15 mgEAA/mL. However, this extract (EtOH-H2O) gave the weakest activity with radical scavenging activity (23.73 ± 0.11 µg/mL). The best anti-DPPH* is obtained with acetone extract (1.93 ± 0.05 µg/mL). Concerning anti-IRP activity, hexane fraction gave the best result with 0.12 ± 0.00 µg/mL. At the antiplasmodial level, the best percentage reduction (19.9 %) is obtained with a dose of 150 mg/kg. On the phytochemistry plan, aqueous and ethyl acetate fractions given the best content with respectively 119.14 ± 8.13 mg EAG/g for total phenolic and 5.33 ± 0.17 EQ/g for total flavonoids. Mineral content quantification show that this plant is rich in calcium with 23.43 ± 1.32 mmol/L. The results due to the antiplasmodial activity could justify the traditional use of this plant.

Aims: The purpose of the study was to determine the antioxidant activity, quantify total phenols and total flavonoids and characterize the secondary metabolites present in methanolic extracts of Chamaecrista hildebrandtii and Clerodendrum rotundifolium using liquid chromatography coupled to mass spectrometry (LC-MS). Methodology: The total phenol and flavonoid contents were determined spectrophotometrically while the antioxidant activity was evaluated using the 2, 2-Diphenyl-1-Picrylhydrazyl (DPPH) free radical scavenging method. The secondary metabolites present in the methanolic leaves extracts were evaluated using LC-MS. Results: The extracts of C. hildebrandtii showed a significantly higher antioxidant activity (IC50 = 8.7 mg/mL) compared to C. rotundifolium (IC50= 28.5 mg/mL). Both methanolic extracts of C. hildebrandtii and C. rotundifolium had common and different types of flavonoids such as quercetin, rutin, (+)-catechin 3-O-gallate and luteolin 6-C-glucoside among others that could be responsible for the observed antioxidant activity. The total phenolic content of C. hildebrandtii (1.33±0.07 mg/g tannic acid equivalents) was significantly higher than that of C. rotundifolium (0.25±0.00 mg/g tannic acid equivalents). However, there was no statistically significant difference (p>0.05) in total flavonoid content of C. hildebrandtii (2.69±0.33 mg/g catechin equivalents) and C. rotundifolium (2.36±0.16 mg/g catechin equivalents). Conclusion: The results of the present study suggested that the good antioxidant activity exhibited by C. hildebrandtii may probably have been brought about by various secondary metabolites functioning in synergy.
