Antinociceptive activity of ethanolic extract of Azadirachta indica A. Juss (Neem, Meliaceae) fruit through opioid, glutamatergic and acid-sensitive ion pathways in adult zebrafish (Danio rerio)

Abstract: Neem fruit (Azadirachta indica A. Juss.) are popularly used to treat infections, diarrhea, fever, bronchitis, skin diseases, infected burns and hypertension. Although the antinociceptive and anti-inflammatory potential of A. indica has already been investigated in experimental models of pain and inflammation in mice, the current research is the first to report the evaluation of the capacity of A. indica fruit ethanolic extract (EtFrNeem) in acute pain attenuation using the adult zebrafish (Danio rerio) as an a… Show more

“…Immediately after behavioral tests, zebrafish were euthanized (10 min immersion in ice water, 2-4 • C) until loss of opercular motions [56], and their whole brains were isolated for biochemical parameters assay. The brains were gently homogenized in ice 0.1 M potassium phosphate buffer (pH 7.4), 1.15% KCl with Potter Homogenizer (Cole-Parmer, Vermon Hills, IL, USA).…”

“…All zebrafish were euthanized (10 min immersion in ice water, 2–4 °C) until loss of opercular motions [ 23 ], and their whole brains were isolated for a biochemical parameters assay. The brains were gently homogenized in ice 0.1 M potassium phosphate buffer (pH 7.4), 1.15% KCl with Potter Homogenizer (Cole-Parmer, Vermon Hills, IL, USA).…”

“…At the end of the experiment, all fish were euthanized by immersion for 10 min in ice water (2–4°C) until the disappearance of opercular movements [33]. The brain samples were dissected out in the ice.…”
