Abstract: Despite the arsenal of technologies employed to control foodborne nontyphoidal Salmonella (NTS), infections have not declined in decades. Poultry is the primary source of NTS outbreaks, as well as the fastest growing meat sector worldwide. With recent FDA rules for phasing-out antibiotics in animal production, pressure is mounting to develop new pathogen reduction strategies. We report on a technology to reduce Salmonella enteritidis in poultry. We engineered probiotic E. coli Nissle 1917, to express and secre… Show more

“…44 Inflammatory responses regulating proinflammatory cytokine production and gene expression in the intestine ultimately result in IBD. 28,29,50 Furthermore, our previous study also indicated that MccJ25 in vitro exerts strong antimicrobial activity against gram-negative bacteria, especially ETEC K88, and that it poses no risk of toxicity for hosts and cells. It is confirmed that ETEC infection activates the inflammatory responses of intestinal epithelial cells.…”

“…49 Several published studies have shown that natural or engineered MccJ25 administration can decrease Salmonella pathogen numbers in mouse and turkey models. 28,29,50 Furthermore, our previous study also indicated that MccJ25 in vitro exerts strong antimicrobial activity against gram-negative bacteria, especially ETEC K88, and that it poses no risk of toxicity for hosts and cells. 8,30 In in vivo experiments, MccJ25 dietary supplements in the form of feed additives for pigs, as well as oral administration of MccJ25, can decrease E. coli numbers in feces.…”

“…Over 3 900 natural and 1 600 synthetic AMPs have been reported, although several show promising therapeutic effects, there are still challenges to their applications, including non‐native structure in some environments, lack of selectivity towards specific pathogen, large‐scale production and delivery to the target position, and reduction in activity due to surface coating . Several approaches have been suggested to develop more effective AMPs, including modification with covalent bonds, sequence changes, addition of amide group, insertion of unnatural amino acid, and computer modeling to design more effective peptides .…”

“…and probiotic Escherichia coli Nissle 1917 [1,14,15,16], have been engineered to produce specific AMPs. In our previous work, we demonstrated the expression of AMPs and their secretion in inhibitory amounts against various pathogens, including Enterococcus faecalis, Enterococcus faecium [1,14], Escherichia coli [15], and Salmonella enterica [16]. …”
