The study was undertaken to isolate, screen, and identify actinomycetes with antimicrobial metabolites. Twenty-one composite soil samples were randomly collected from various unique agroecological niches in the Rift Valley of Ethiopia. The soil samples were serially diluted and spread on starch casein agar medium supplemented with 50 μg/ml cycloheximide and 25 μg/ml nalidixic acid. Two hundred and forty-nine (249) actinomycetes cultures were isolated and screened by cross streaking against various human pathogens. Twenty-four isolates with pronounced antimicrobial activity were selected for identification and further screening. Among the isolates, 172 (69.1%) showed antimicrobial activities against tested pathogens. The inhibition zone of the isolates ranged from 5 ± 0.31 to >40 mm during primary screening. The antimicrobial activity of the crude extracts of promising isolates showed a statistically significant difference P < 0.05 between them and the control. The isolates RVE129 and RVE217 showed the maximum zone of inhibition at 27 ± 0.6 mm and 26 ± 0.6 mm, respectively, against S. aureus, and the results were higher than the standard drug streptomycin (25 ± 0.58 mm). The inhibition zone of crude extracts from RVE129 was at the maximum of 22 ± 0.0 mm against P . aeruginosa, almost comparable to the standard drug streptomycin (24 ± 0.58 mm). Crude extract from the isolates RVE129 and RVE187 showed higher inhibition zones of 22 ± 0.6 mm and 16 ± 0.33 mm against A. niger ATCC10535, which, however, were smaller than those obtained with the standard drug amphotericin B (29 ± 0.6 mm). Twenty-four actinomycete strains with remarkable bioactivity were characterized using various cultural, morphological, physiological, and biochemical characteristics and assigned under the genus Streptomycetes. The finding of the current study indicates that Streptomyces sp. isolated from the Rift Valley of Ethiopia was found to possess a broad spectrum of bioactivity against a range of human pathogens.

Background Actinomycetes are prolific producers of bioactive compounds which can be used to combat microbial infections. Microbial infections have resulted to increase in mortality and morbidity rates world-wide, especially in developing nations. This study was carried out to isolate and identify actinomycetes with potentials of synthesizing bioactive compounds, to produce and partially purify antimicrobial extracts, to characterize bioactive compounds in the extracts, to assess their antibacterial activity and to profile other biological properties of the bioactive compounds synthesized by the actinomycetes. Results Streptomyces sp. SUI (MT584797), Streptomyces sp. SW72IV (MT584818) and Streptomyces sp. SW72VII (MT584816) were isolated and identified. Ultraviolet–visible spectra absorption ranged between 241 and 251 nm wavelengths showing the polyene or chromone and unsaturation nature of the natural compounds. Infrared (IR) spectra indicated functional groups such as hydroxyl, aliphatic bromo, carbonyls, esters, carboxylic acids and silicon oxy compounds in the compounds produced by the three strains. Gas Chromatography-Mass Spectrum (GC–MS) identified elaidic acid isopropyl ester (32.11%), Octadec-9-enoic acid (17.44%) and 2, 3-dihydroxyl elaidate (10%) to be mostly produced by Streptomyces sp. SUI, Streptomyces sp. SW72IV and Streptomyces sp. SW72VII respectively. The three strains exhibited antimicrobial activity against Bacillus sp. Pseudomonas aeruginosa ATCC 9077, Staphylococcus aureus ATCC 700699, Candida albicans and Aspergilus flavus. Conclusion The results showed that the three strains of Streptomyces could be sources of antimicrobial bioactive compounds and other secondary metabolites that can be used in the production of pharmaceutical bioactive agents that are effective against pathogens, and production of biological materials that can be used in cosmetics and food industries.

Here, we reported the biosynthesis of silver nanoparticles (AgNPs) using Urtica dioica (nettle) leaf extract as green reducing and capping agents and investigate their anticancer and antibacterial, activity. The Nettle‐mediated biosynthesized AgNPs was characterized by UV–Vis a spectrophotometer. Their size, shape and elemental analysis were determined with the using of SEM and TEM. The crystal structure was determined by XRD and the biomolecules responsible for the reduction of Ag+ were determined using FTIR analysis. Nettle‐mediated biosynthesis AgNPs indicated strong antibacterial activity against pathogenic microorganisms. Again, the antioxidant activity of AgNPs is quite high when compared to ascorbic acid. Anticancer effect of AgNPs, IC50 dose was determined by XTT analysis using MCF‐7 cell line and the IC50 value was found to be 0.243 ± 0.014 μg/mL (% w/v).
