Antibody neutralization to SARS-CoV-2 and variants after 1 year in Wuhan, China

Abstract: Sera from a large cohort of COVID-19 convalescents in Wuhan were collected for evaluation of anti-SARS-CoV-2 humoral immunity -Anti-SARS-CoV-2 IgG was well maintained for 1 year in most convalescents and can potently neutralize the original strain and the B.1.1.7 variant -Varying degrees of immune escape was observed on the tested variants, especially on B.1.351 and B.1.617.2 variants -Sera from a few individuals showed remarkably broad neutralization potency against SARS-CoV-2 WT and variants ll www.cell.com/… Show more

“…By contrast, the fluorescent QD-peptides biosensors could also achieve over 92% sensitivity (vs. 78% sensitivity for ELISA) in authentic COVID-19 serum samples at 1:2000 serum dilution and avoid cross-reactions as much as possible by using SARS-CoV-2 specific B-cells epitopes. Our results are similar to the study by Liu et al (using overlapping sera) finding that high levels of specific antibodies were present 1 year after SARS-CoV-2 infection [ 27 ]. Still, our QD-peptides biosensors (98.1% sensitivity for QD-S epi1 at 1:2000 sera dilution) are more sensitive to the ELISA assay (RBD-based) at an even lower sera dilution (1:100) used in the study by Liu et al [ 27 ].…”

“…These data would have been useful to evaluate the extent and duration of the immune response against SARS-CoV-2. However, the anti-SARS-CoV-2 humoral immunity seems to last for at least 1 year in most convalescents, protecting them from the original virus, as reported by Liu et al [50]. Considering that most subjects were from Modena province, the reliability of seroprevalence estimates stratified by province may be low, especially considering the small sample size.…”

“…Currently, a greatly increasing concern is the emergence of numerous SARS-CoV-2 variants resulting from the rapid spread and constant evolution of virus. In contrast to the potent neutralization against the WT SARS-CoV-2, varying degrees of immune escape by various variants, such as Beta (B.1.351), Delta (B.1.617.2), and Mu (B.1.621), have been observed in recovered human sera after one year [ 8 , 25 ]. In particular, with the emergence of various Omicron variants (BA.1, BA.2, and BA.4/5, etc.…”
