This study presents modeling and optimization of ultrasound-assisted extraction (UAE) of Melastoma malabathricum with the objective of evaluating its phytochemical properties. This one-factor-at-a-time (OFAT) procedure was conducted to screen for optimization variables whose domains included extraction temperature (XET), ultrasonic time (XUT), solvent concentration (XSC), and sample-to-liquid ratio (XSLR). Response surface methodology (RSM) coupled with Box–Behnken design (BBD) was applied to establish optimum conditions for maximum antioxidant extraction. Modeling and optimization conditions of UAE at 37 kHz, XET 32 °C for XUT 16 min and dissolved in an XSC 70% ethanol concentration at a XSLR 1:10 ratio yielded scavenging effects on 2,2-diphenyl-1-picryl-hydrazyl (DPPH) at 96% ± 1.48 and recorded values of total phenolic content (TPC) and total flavonoid content (TFC) at 803.456 ± 32.48 mg GAE (gallic acid equivalents)/g, and 102.972 ± 2.51 mg QE (quercetin equivalents)/g, respectively. The presence of high flavonoid compounds was verified using TWIMS-QTOFMS. Chromatic evaluation of phytochemicals using gas chromatography–mass spectrometry (GC–MS) revealed the presence of 14 phytocompounds widely documented to play significant roles in human health. This study provides a comparative evaluation with other studies and may be used for validation of the species’ potential for its much-acclaimed medicinal and cosmeceutical uses.

Solid-Phase Extraction and Characterization of Quercetrin-Rich Fraction from Melastoma malabathricum Leaves

This study was focused on the recovery of quercetrin from the crude extract of Melastoma malabathricum leaves using the technique of solid-phase extraction. The process variables and their ranges were screened using one-factor-at-a-time and statistically optimized using the response surface methodology. The results found that 9.13 mg/mL of crude extract required 18.24 mL of 70.5% methanol as the eluent to yield an 86.6% w/w fraction containing 36.02 mg/g of quercetrin. The process increased quercetrin from 1% w/w in the crude extract to 4% w/w in the fraction. Quercetrin was likely to be the compound contributing to antiradical and antidiabetic activities. In silico simulation showed that quercetrin had low binding energy and mostly bound with charged (Glu and Arg) and aromatic (Tyr and Phe) amino acids via hydrogen bonds. Its inhibitory progress against DPP-IV was faster than crude extract at low concentration (<100 µg/mL).

Burns are tissue damage caused by contact with heat sources such as fire, chemicals, electricity and radiation. This study aims to find a new burn drug from senduduk leaves (Melastoma malabathricum L.) with a form of standard cream preparation type M/A. The viscous extract of the leaves of senduduk is carried out by a characterization test and screening test, then formulated in the form of cream with a concentration of 2.5%, 5%, and 7.5%. Standard cream type M/A formulated with senduduk leaf extract was evaluated and tested for the stability of the preparation. Mice were burnt with a heated metal plate, then smeared with the cream of senduduk leaf extract and the diameter was measured using a calliper by measuring the four sides of the wound diameter and averaging it to obtain the proportion of wound closure. The percentage of wound closure was analyzed at D-1 to D-11 treatment. The results showed that the ethanol extract of senduduk leaf cream preparation with a concentration of 7.5% was more effective in healing burns with an average diameter of 0.20 cm of final burns and the proportion of wound healing was 99% compared to the concentration of senduduk leaf extract cream preparation which was other. The results of the burn test showed that the ethanol extract of senduduk leaf cream with a concentration of 7.5% was efficacious in healing wounds in mice.
