Introduction: Pain is managed using conventional drugs like paracetamol, aspirin and diclofenac among others. Synthetic drugs have many side effects. This study aimed at evaluating the analgesic potential of the dichloromethane leaf extracts of Eucalyptus globulus and Senna didymobotrya in mice. Methods: The dichloromethane leaf extracts of E. globulus and S. didymobotrya were subjected to quantitative phytochemical analysis using gas chromatography-mass spectrophotometry (GC-MS). In vivo analgesic evaluation comprised of nine groups of animals (Swiss albino mice): normal, positive, negative control and six experimental groups that received 25, 50, 100, 150, 200 and 250 mg/kg body weight of each plant extract intraperitoneally. Thirty minutes later, they were injected with 0.01 mL of 2.5% formalin. The animals in positive control group were administered diclofenac (15 mg/kg) and formalin, the normal control mice received 3% dimethyl sulfoxide (DMSO) in normal saline, while the negative group received DMSO in normal saline and formalin. All the doses were administered intraperitoneally. The duration of shaking and licking of the injected paw was scored and analyzed. Results: The analysis revealed that E. globulus contained alpha-pinenes, endo-fenchol, α-eudesmol, myrcene, camphene, alpha-phellandrene, limonene, and camphor while S. dymobotrya possessed camphene, alpha-phellandrene, limonene, and camphor. In the late phase, E. globulus at the doses of 25, 50, 100, 150, 200 and 250 mg/kg reduced the paw licking time by 34.03%, 60.79%, 84.33 %, 90.65%, 94.49%, 98.52%, respectively while S. didymobotrya extract reduced the paw licking time by 26.48%, 32.96%, 87.04%, 91.27%, 93.40%, 90.97%, and 96.82%, respectively. Conclusion: The results of this study validate and support the traditional uses of these plants as analgesics.

Introduction. Diarrheal diseases are associated with an estimated 1.3 million deaths annually, with most occurring in resource-limited countries; up to 25% of deaths in young children living in Africa and southeast Asia are attributable to acute gastroenteritis. Due to limitations associated with various treatments available, the need for developing newer drugs is imperative. Objective. This study was aimed to evaluate the antidiarrheal activity of root extract and fractions of C. abyssinica Jaub. & Spach. (Euphorbiaceae) in mice. Methods. After plant extraction and subsequent fractionation of the crude extract, the antidiarrheal activity was screened in castor oil induced diarrhea, castor oil induced enteropooling, and gastrointestinal motility test models accordingly. Result. The root extract of C. abyssinica produced neither visible signs of toxicity nor death at a single dose of 2000 mg/kg, suggesting the LD50 > 2000 mg/kg. In the castor oil induced diarrheal model, the highest dose of the extract (400 mg/kg) showed a maximal inhibition in the onset (158.00 ± 14.64, p<0.01, in minutes) of wet feces as compared to the negative control. In the enteropooling model, 400 mg/kg treated mice showed a significant reduction in volume (0.47 ± 0.02 ml, p<0.01) and weight (0.50 ± 0.02 g, p<0.05) of intestinal content as compared to the vehicle treated group. In the gastrointestinal motility test, the hydromethanolic root extract of C. abyssinica significantly inhibited the intestinal transit of charcoal meal at 400 mg/kg. In addition, chloroform and n-butanol fractions significantly reduced the distance moved by charcoal at doses of 200 mg/kg and 400 mg/kg, whereas aqueous fraction showed a significant effect at all test doses. The highest antidiarrheal index was observed at the maximal dose of extract and each fraction. Conclusion. The results obtained showed that the findings provide scientific support for the folkloric repute of C. abyssinica roots as treatment of diarrhea.

Global use of Ethnomedicinal Plants to Treat Toothache

Toothache is one of the most common global health problems, and medicinal plants are widely used to relieve the associated pain and inflammation. Several studies have been conducted on the use of plants to treat toothache, but no study has comprehensively assessed the types of plants and the mechanisms of action of the phytochemical compounds involved in their analgesic effect. This review aims to bridge this gap. This is the first review to collect a large volume of data on the global use of medicinal plants used in the treatment of toothache. It presents the relevant information for dentists, researchers, and academics on using medicinal plants to treat toothache. We found that preclinical studies and state-of-the-art technology hold promise for furthering our knowledge of this important topic. In total, 21 species of medicinal plants used to treat toothache were found in America, 29 in Europe, 192 in Africa, 112 in Asia, and 10 in Oceania. The most common species were Allium sativum, Allium cepa, Acmella oleracea, Jatropha curcas, Jatropha gossypiifolia, and Syzygium aromaticum. The most commonly found family of medicinal plants was Asteraceae, followed by Solanaceae, Fabaceae, Lamiaceae, Euphorbiaceae, Rutaceae, and Myrtaceae. The most common phytochemicals found were flavonoids, terpenes, polyphenols, and alkaloids. The reported mechanisms of action involved in toothache analgesia were antioxidant effects, effects mediated by transient receptor potential channels, the γ-aminobutyric acid mechanism, and the cyclooxygenase/lipoxygenase anti-inflammatory mechanism.
