Anti-Mycobacterium tuberculosis activity of antituberculosis drugs and amoxicillin/clavulanate combination

Abstract: We report the in vitro drugs interaction by the resazurin drugs combination microtiter assay (REDCA) of amoxicillin (AMO)/clavulanate (CLAV) with isoniazid (INH), ethambutol (EMB), and rifampicin (RIF) against susceptible and resistant Mycobacterium tuberculosis isolates. The addition of AMO/CLAV to classical antituberculosis drugs should be explored as a promising alternative for the treatment of resistant tuberculosis (TB).

“…The in vitro growth interaction of bacteria with the inhibitors and the first line drugs such as rifampicin (RIF)/INH was determined by resazurin drugs combination microtiter assay [ 33 ]. Two-fold serial dilutions ranging from 12.5 mg/L to 0.04 mg/L for RIF/INH were combined with the same concentration of inhibitor (ranging from 12.5 mg/L to 0.04 mg/L) in wells of microtiter plates followed by the addition of bacteria and resazurin solution as described above.…”

“…The effects of the drug combinations were classified as synergistic (FICI ≤ 0.5), additive or no interaction (FICI > 0.5–4), and antagonistic (FICI > 4) as described by Pagliotto et al [ 33 ].…”

“…Furthermore, the ␤-lactamase inhibitor clavulanate as a component of triple combinations including cephalosporins (and other beta-lactams) proved to be a key synergistic partner due to its ability to rescue the activity of RIF against a RIF-resistant strain (2). A combination of amoxicillin-clavulanate with first-line anti-TB drugs has also been explored and showed potent synergism against MDR isolates (4). Repurposing of drugs such as spectinomycin was also studied to test the concept that clinically available antibiotics with limited efficacy against M. tuberculosis might be used for TB treatment when coadministered with macrolides and azoles (5).…”

“…Beta-lactams are reported to have early antitubercular activity comparable to the conventional ATT drugs other than isoniazid. 17 We hypothesize these reasons for Xpert positive culture negative results.…”
