Anti–Monocyte Chemoattractant Protein-1/Monocyte Chemotactic and Activating Factor Antibody Inhibits Neointimal Hyperplasia in Injured Rat Carotid Arteries

Abstract: Abstract-Monocyte chemoattractant protein-1 (MCP-1)/monocyte chemotactic and activating factor (MCAF) has been suggested to promote atherogenesis. The effects of in vivo neutralization of MCP-1 in a rat model were examined in an effort to clarify the role of MCP-1 in the development of neointimal hyperplasia. Competitive polymerase chain reaction analysis revealed maximum MCP-1 mRNA expression at 4 hours after carotid arterial injury. Increased immunoreactivities of MCP-1 were also detected at 2 and 8 hours af… Show more

“…Non-treated VSMCs or VSMCs treated with function blocking mAbs for 1 h were then incubated at room temperature for 1 h. After washing, adherent cells were fixed with 1% glutaraldehyde, stained with crystal violet, and quantified by dye extraction with the measurement of absorbance at 595nm. VSMCs migration was measured with the transwell migration assay (Furukawa et al, 1999). In brief, VSMCs were added into the upper chamber of a seeded transwell containing serum-free medium, while the lower chamber was supplemented with serum-free medium containing the indicated recombinant CCN4 or PDGF-BB as a positive control.…”

“…In addition, Okada et al (1998) have reported that MCP-1 is up-regulated by cyclic stretch in human endothelial cells, and Furukawa et al (1999) have reported that anti-MCP-1 treatment reduces neointimal hyperplasia in early neointimal lesion formation at balloon injury of rat carotid artery. Accordingly, mod- ulating the production or inhibiting the action of MCP-1 may also lead to the prevention of balloon injury and restenosis, and this may truly be a case of killing two birds with one stone in intervention by catheter for AMI.…”
