Abstract: Major challenges for current therapeutic strategies against breast cancer are associated with drug-induced toxicities. Considering the immense potential of bioactive phytochemicals to deliver non-toxic, efficient anti-cancer therapeutics, we performed bio-guided fractionation of Eclipta alba extract and discovered that particularly the chloroform fraction of Eclipta alba (CFEA) is selectively inducing cytotoxicity to breast cancer cells over non-tumorigenic breast epithelial cells. Our unbiased mechanistic hun… Show more

“…Luteolin was also found to be more active than the twice as available coumarin wedelolactone component of E. alba [89]. Chloroform extracted E. alba luteolin was effective at 50, 100, and 200 μg/ml.…”

“…In vitro and in vivo mouse studies showed that the flavone, chloroform extracted luteolin from Eclipta alba (Bhringraj) dose and time dependently selectively activates intrinsic apoptosis by releasing mitochondrial apoptogenic proteins into the cytosol, inhibiting caspase-9, up regulating pro-apoptotic molecular chaperone heat shock protein 60 (Hsp60) and down regulating anti-apoptotic protein XIAP in MCF-7 and MDA-MB-231 cells, but spares MCF-10A normal breast epithelial cells [89]. Luteolin was also found to be more active than the twice as available coumarin wedelolactone component of E. alba [89].…”

“…The life span of BLEA treated mice was calculated and compared with untreated tumor-bearing animals. The life span of experimental animals was monitored and calculated by using the following formula [(T − C)/C] × 100, where ‘T’ indicates the number of days the treated animals survived and ‘C’ indicates that number of days that tumor animals survived (Arya et al, 2015).…”

“…This method has a several drawback due to involvement of large amount of drug. (Arya et al, 2015). These drugs are often cytotoxic and induce cancer cell death in solid tumors through various route like cell cycle arrest, DNA synthesis inhibition, necrotic cell death, or by inducing apoptosis.…”

“…In vitro cytotoxic activities of Doxorubicin, Paclitaxel, 5-Fluorouracil, Cisplatin, TRAIL, and AMD3100 were assessed by using standard SRB assay as described before [23,24]. The absorbance of the treated and untreated cells was measured on a multi-well scanning spectrophotometer (Epoch Microplate Reader, Biotek, USA) at a wavelength of 510 nm.…”
