Angiotensin-Converting Enzyme 2 (ACE2) in Disease Pathogenesis

Abstract: Angiotensin-converting enzyme 2 (ACE2), a first homolog of ACE, regulates the renin-angiotensin system by counterbalancing ACE activity. Accumulating evidence in recent years has demonstrated a physiological and pathological role of ACE2 in the cardiovascular, renal and respiratory systems. For instance, in the acute respiratory distress syndrome (ARDS), ACE, AngII, and AT1R promote the disease pathogenesis, whereas ACE2 and the AT2R protect from ARDS. Importantly, ACE2 has been identified as a key SARS-corona… Show more

“…This polymorphism was later classified as SNP rs4646994, and was shown to have a considerable influence on the level of ACE in plasma, where the D allele was associated with an elevated level (16, 18). For over two decades considerable interest had been directed to the study of I/D polymorphism in association with different disease states (19, 20). Several diseases were related to the frequency of I and D alleles and II, ID or DD genotypes of the ACE gene.…”

“…In contrast to angiotensin converting enzyme (ACE), which synthesizes the potent vasoconstrictor, angiotensin II (Ang II) through its dicarboxypeptidase activity acting on the decapeptide angiotensin I, ACE2 catabolizes Ang II through its monocarboxypeptidase activity (Figure 1). Studies suggest ACE2 counters the adverse effects of ACE action and that these two enzymes play a ying and yang role in the physiological regulation of Ang II metabolism and in disease pathologies such as hypertension, progressive renal disease and diabetes [1,4,5]. …”

“…Angiotensin-converting enzyme 2 (ACE2), a homolog of ACE, is a carboxypeptidase that degrades angiotensin (Ang) II to Ang-(1-7) [3,4]. ACE2 plays an important role in the vasoprotective axis (ACE2-Ang-(1-7)-Mas axis) of the renin angiotensin system (RAS) and counterbalances the vasoconstrictive, proliferative and fibrotic axes [ACE-Ang II-Ang II type 1 receptor (AT1R) axis] of the RAS [5,6,7].…”
