Background/Aims: Cardiovascular complications are common in astronauts returning from a prolonged spaceflight. These health problems might be driven by complex modulations of gene expression and protein synthesis in endothelial cells (ECs). Studies on the influence of microgravity on phenotype, growth pattern and biological processes of ECs can help to understand these complications. Methods: We exposed ECs (EA.hy926) to a Random Positioning Machine (RPM). Proteins associated with cell structure, angiogenesis and endothelial dysfunction were investigated in distinct pools of multicellular spheroids (MCS), adherent cells (AD) and tubular structures (TS) formed after a 35-day RPM-exposure. Results: Combining morphological and molecular approaches, we found AD, MCS and TS with changes in the synthesis and release of proteins involved in three-dimensional growth. Fibronectin and monocyte chemoattractant protein-1 (MCP-1) mRNAs and protein contents were elevated along with an increased secretion of vascular endothelial growth factor (VEGF), interleukin (IL)-6, IL-8, MCP-1, intercellular adhesion molecule 1 (ICAM-1), vascular cell adhesion molecule 1 (VCAM-1), neutrophil gelatinase-associated lipocalin (NGAL) and regulated on activation, normal T cell expressed and secreted (RANTES) proteins in the culture supernatant as determined by multianalyte profiling technology. Together they form a network of interaction. Conclusions: These results show that a prolonged RPM-exposure of ECs induced TS and MCS formation. The factors VEGF, NGAL, IL-6, IL-8, MCP-1, VCAM-1, ICAM-1, fibronectin and RANTES seem to be affected when gravity is omitted.

Angiopoietin (Ang)-2, a ligand of the receptor tyrosine kinase Tie2, is known to be involved in the regulation of embryonic lymphangiogenesis. However, the role of Ang-2 in postnatal pathological lymphangiogenesis, such as inflammation, is largely unknown. We used a combination of imaging, molecular, and cellular approaches to investigate whether Ang-2 is involved in inflammatory lymphangiogenesis. We observed strong and continuous expression of Ang-2 on newly generated lymphatic vessels for 2 wk in sutured corneas of BALB/c mice. This expression was concurrent with an increased number of lymphatic vessels. TNF-α expression also increased, with peak TNF-α expression occurring before peak Ang-2 expression was reached. In vitro experiments showed that TNF-α stimulates Ang-2 and Tie2 and ICAM-1 expression on human lymphatic endothelial cells (LECs) and blood vascular endothelial cells (BECs). Ang-2 alone did not affect the biological behavior of LECs, whereas Ang-2 combined with TNF-α significantly promoted the proliferation of LECs but not BECs. In mouse models, blockade of Ang-2 with L1-10, an Ang-2-specific inhibitor, significantly inhibited lymphangiogenesis but promoted angiogenesis. These results clearly indicate that Ang-2 acts as a crucial regulator of inflammatory lymphangiogenesis by sensitizing the lymphatic vasculature to inflammatory stimuli, thereby directly promoting lymphangiogenesis. The involvement of Ang-2 in inflammatory lymphangiogenesis provides a strong rationale for the exploitation of anti-Ang-2 treatment in the prevention and treatment of tumor metastasis and transplant rejection.

External Financing Efficiency of Rural Revitalization Listed Companies in China—Based on Two-Stage DEA and Grey Relational Analysis

Rural revitalization is an important strategy to promote sustainable development of rural areas in China. Rural revitalization listed companies play an important role in implementing the rural revitalization strategy and developing the agricultural industry. However, the financing problem has always been a bottleneck problem with Chinese listed companies. This study used a two-stage DEA (data envelopment analysis) method to evaluate the funds raising efficiency, funds using efficiency, and overall financing efficiency of 34 rural revitalization listed companies in 2018. The results show that the financing efficiencies of 34 sample companies were low, only six companies have overall efficient financing, and there was much room for improvement. Financing efficiency varied greatly depending on the nature of the company, the industry, and the listing board. The efficiency of funds using of state-owned enterprises was much lower than that of private companies. The average efficiency value of agricultural company funds raising was lower than that of manufacturing. The efficiency of small and middle-size enterprises (SMEs) was lower than that of main board companies, but the growth enterprise market (GEM) companies achieved higher efficiency in the funds using. Further, by using the grey relational analysis (GRA) method, we found that the key factors affecting the financing efficiency of sample companies included capital structure, debt-paying ability, governance structure, company age, and operating ability. To improve financing efficiency, the companies should not only optimize their capital structure and governance structure but also improve their management and innovation capabilities. At the same time, the state also needs to give different policies support according to the characteristics of the companies.
