Analysis of spermidine and spermine in rat liver. Effect of hypophysectomy on polyamine concentrations

Abstract: A simple, rapid, reproducible assay for tissue polyamines using a standard unmodified amino-acid analyzer is described. This technique will allow accurate measurement of the polyamines from as little as 10 mg of liver in less than 1 h. We have used this method to establish the optimum conditions for the extraction of polyamines from liver and to study the effects of hypophysectomy on polyamine content of rat liver. Hypophysectomy caused a fall in RNA, protein, putresine, and spermidine content prevented the no… Show more

“…For analysis of polyamine concentrations, a portion of homogenate was deproteinized by addition of solid sulphosalicylic acid (40mg/ml of homogenate), protein was removed by centrifugation (20000g for 10min at 4°C), and the clear supernatant was applied to a Beckman amino acid analyser, as described previously (Hall et al, 1978).…”

“…3. Putrescine concentration and ratio of activity of ornithine decarboxylase to S-adenosylmethionine decarboxylase in rat liver during and after weaning Putrescine (a) was measured on a Beckman model 121 amino acid analyser (Hall et al, 1978). Each point is the mean ± S.E.M.…”

“…In particular, the reported effects of spermidine include promoting protein synthesis, anti-inflammation, antioxidation, and promoting hair growth. [34][35][36][37] In terms of elevated SAM levels, the significance of OM-X ® administration is expected to be not only activation of detoxification but also promotion of protein synthesis and anti-aging effects of spermidine.…”
