Analgesic efficacy of single oral doses of lumiracoxib and ibuprofen in patients with postoperative dental pain

Abstract: This randomised, double-blind study compared single dose lumiracoxib (a cyclooxygenase-2 selective inhibitor) 100 and 400 mg, ibuprofen 400 mg and placebo in patients with postoperative dental pain over 12 h. The primary efficacy variable was pain intensity difference. Lumiracoxib 400 mg and ibuprofen were superior to placebo from 1 to 12 h post dose while lumiracoxib 100 mg was superior from 1.5 to 9 h. Lumiracoxib 400 mg demonstrated the fastest median time to onset of analgesia (37.4 min) followed by ibupro… Show more

“…3 Acute dental pain and postoperative pain are often treated with analgesics and non-steroidal anti-inflammatory drugs (NSAIDs) such as paracetamol, ibuprofen, rofecoxib and celecoxib. [4][5][6] The efficacy of these drugs in reducing inflammation and pain is largely attributed to prevention of prostaglandin synthesis via non-specific inhibition of both cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2). 5 Lumiracoxib is a new selective inhibitor of COX-2 that was also developed for treating acute pain.…”

“…6 The second one compared lumiracoxib 400 mg with rofecoxib 50 mg, celecoxib 200 mg and placebo, and 5 The third compared lumiracoxib 400 mg with celecoxib 400 mg and placebo, and assessed 364 patients. 4 These studies were all carried out in the United States with financial support from the pharmaceutical industry.…”

“…Thrombocytopenia, cutaneous eruptions, visual disturbance, fluid retention, and edema occur less frequently. 4 The aim of the present study was to assess the onset of the analgesic effect and the effectiveness and tolerability of oral treatment with a single dose of nimesulide 300 mg versus ibuprofen 400 mg q6h over 24 hours in patients with moderate or severe pain after surgical extraction of an impacted third molar.…”

“…This type of pain is widely used to evaluate the effectiveness of analgesic agents, especially in single doses. [1][2][3][4] Oral nonsteroidal anti-inflammatory drugs (NSAIDs) are effective in relieving mild or moderate pain, and they are more likely to be used in ambulatory patients compared with their injectable counterparts. The analgesic action of nimesulide has been documented in a wide range of acute and chronic pain.…”
