An UPLC-MS/MS Method for the Simultaneous Identification and Quantitation of Cell Wall Phenolics in Brassica napus Seeds

Abstract: The seed residues left after pressing of rapeseed oil are rich in proteins and could be used for human nutrition and animal feeding. These press cakes contain, however, antinutritives, with fiber being the most abundant one. The analysis of fiber phenolic component (localized to seed coat cell walls) is, therefore, important in breeding and food quality control. However, correct structure and content assignments of cell wall-bound phenolics are challenging due to their low stability during sample preparation. … Show more

“…Phenolic compounds play diverse roles in plants. These compounds provide structural support of the cell wall or protect plants against pathogens, herbivores, and UV radiation [17]. Of all classes of secondary metabolites, phenolics, specifically flavonoids, are the most relevant for UV protection.…”

“…Due to their cis-diol groups, these metabolites saturate the binding sites of the affinity column and reduce the peptide enrichment efficiency. Moreover, phenylpropanoids, as well as their sugar esters and glycosides, are well-retained on reversed phase [63] and might co-elute with peptides in course of RP-HPLC separation, leading to essential ion suppression during the ESI process and shift of peptide signals in the lower part on the instrument dynamic range. Thus, the analysis of early glycation proteins in total extracts that can not be efficiently ultrafiltrated is a challenging task.…”

“…Rapeseed ( Brassica napus; Cruciferae ) is an important oil crop in agriculture worldwide, and ranks the third largest source of vegetable oil all over the world [1,2,3]. Traditional rapeseed oil contains more than 40% erucic acid [4], which could pose a risk to human health when excessive amounts are consumed [5].…”

“…Metabolomics was used to investigate the chemical composition of different organs of Brassica napus L. using UPLC-QTOF-PDAP-MS [17]. The composition of oilseed rape and turnip rape seeds has been investigated by nuclear magnetic resonance (NMR) [18] and the cell wall phenolics by UPLC-MS/MS [1]. Comparative metabolomics was employed to evaluate Chinese and North American rice [16], analyze carrots from different agronomic environments [19], distinguish Zingiber officinale Roscoe from two geographical origins [20], expound the profile of fruits from Tanzania [15], and characterize the metabolic changes of cultivated and wild soybean under salt stress [21].…”
