An Integrative Pharmacology Based Analysis of Refined Liuweiwuling Against Liver Injury: A Novel Component Combination and Hepaprotective Mechanism

Abstract: Liver disease is a major cause of illness and death worldwide. In China, liver diseases, primarily alcoholic and nonalcoholic fatty liver disease, and viral hepatitis, affect approximately 300 million people, resulting in a major impact on the global burden of liver diseases. The use of Liuweiwuling (LWWL), a traditional Chinese medicine formula, approved by the Chinese Food and Drug Administration for decreasing aminotransferase levels induced by different liver diseases. Our previous study indicated a part o… Show more

“…IκBα was phosphorylated and subsequently contributed to the phosphorylation of NF-κB [62] . NF-κB activation can result in the production of pro-inflammatory cytokines, and the polarization of M1 macrophages, which can ultimately create severe liver damage [36 , [63] , [64] , [65] . Consequently, suppressing NF-κB activation is an encouraging strategy for treating ALI [66] .…”

“…LWWL, as a drug used clinically in combination with NAs to treat hepatitis B, has hepatoprotective and enzyme-lowering effects, as well as antiviral effects in vitro and in vivo [ 26 , 40 ]. Building on this foundation, we further confirmed the antiviral effect of LWWL in hepatitis B by observing reductions in the level of HBV DNA, HBsAg, and HBeAg in serum.…”

“…Scavenging ROS effects of esculetin. Accumulation of ROS can activate the mitochondrial apoptotic pathway and induce apoptosis, and esculetin can block this process by scavenging ROS (32). In zinc oxide nanoparticles-induced human neuroblastoma SH-SY5Y cells, esculetin (10-30 µM) can decrease GPx and GSH depletion by inhibiting lipoxygenase expression, which ameliorates the ROS accumulation-induced apoptosis (33).…”
