Dynamic and adaptive materials are powerful constructs in macromolecular and polymer chemistry with a wide array of applications in drug delivery, bioactive systems, and self‐healing materials. Very often, dynamic materials are based on carefully tailored cyclodextrin host–guest interactions. The precise incorporation of these host and guest moieties into macromolecular building blocks allows the formation of complex macromolecular structures with predefined functions. Thus, dynamic materials with extraordinary adaptive property profiles—responsive to thermal, chemical, and photonic fields—become accessible. This Review explores the hierarchical formation of dynamic materials and complex macromolecular structures from the molecular via the macromolecular to the colloidal and macroscopic level, with a specific emphasis on the functionality and responsiveness of the assemblies, specifically in biological contexts.

“…Dextran (3.00 g) was dissolved in deionized water (100 mL) at 50°C. After cooling to the room temperature, sodium periodate (3.96 g, 18.52 m M ) was added to the solution . The reaction was conducted at 25°C for 2 h. Then ethylene glycol (1.15 g, 18.52 m M ) was added to the solution proceeding for 2 h. The solution was dialyzed against water for 48 h by using a dialysis bag MD44 (MWCO: 8000‐14,000, America) to remove the by‐products and then freeze‐dried (yield: 2.79 g, 93%).…”

A novel voriconazole (VCZ)‐loaded injectable hydrogel was in situ synthetized via a Schiff base reaction between polyaldehyde dextran (PAD) and carboxymethyl chitosan (CMCTS) for intraocular drug loading and release. Water‐insoluble VCZ, which is an effective agent in clinic treating fungal endophthalmitis, was loaded through the inclusion into the β‐cyclodextrin (β‐CD) cavity based on host‐guest interaction on the linear poly β‐CD chain, which was in situ twined in the cross‐linked hydrogel structure. The gelation time, degradation, and drug release process were investigated by studying three kinds of hydrogels with different volume ratio of PAD to CMCTS resolving in phosphate buffered saline (PBS) solution (3:7, 6:7, 9:7), respectively. The property of VCZ‐loaded injectable hydrogel should be adjusted through changing the cross‐linked density of the hydrogel, the molecular weight or concentration of the linear poly β‐CD to meet the treating requirement, and a multistage drug controlled‐release mechanism was proposed. In conclusion, the VCZ‐loaded in situ injectable hydrogel should be offered as a promising strategy for intraocular drug delivery in vitreous cavity for the treatment of fungal endophthalmitis. © 2019 Wiley Periodicals, Inc. Water‐soluble poly(ethylene glycol) derivatives with multiple “clickable” mercapto groups or double bonds were facilely synthesized in a large scale by direct polycondensation of oligo(ethylene glycol) diol with mercaptosuccinic acid or maleic acid catalyzed by scandium trifluoromethanesulfonate under mild conditions. Injectable hydrogels containing doxorubicin hydrochloride (DOX · HCl) could be rapidly formed using these poly(ethylene glycol) derivatives as precursors via in situ thiol‐ene “click” reaction under physiological conditions without light, initiator, or metal catalyst. DOX · HCl could be sustained released from the hydrogels as a result of the hindrance of the three dimensional hydrogel network on the drug molecules, which makes this kind of DOX‐loaded hydrogels a promising candidate for localized tumor chemotherapy.
