An Inhalable Powder Formulation Based on Micro- and Nanoparticles Containing 5-Fluorouracil for the Treatment of Metastatic Melanoma

Abstract: Melanoma is the most aggressive and lethal type of skin cancer, with a poor prognosis because of the potential for metastatic spread. The aim was to develop innovative powder formulations for the treatment of metastatic melanoma based on micro- and nanocarriers containing 5-fluorouracil (5FU) for pulmonary administration, aiming at local and systemic action. Therefore, two innovative inhalable powder formulations were produced by spray-drying using chondroitin sulfate as a structuring polymer: (a) 5FU nanopart… Show more

“…The scanning electron microscopy (SEM) images obtained from the formulations DAP-LNC-CS-L1 and DAP-LNC-CS-L2 showed the presence of morphological agglomerates and an irregular surface of the particles according to Figure 6. The SEM images found in our study corroborate with other dry powder images of spray-dried nanocapsules obtained by SEM (22,23,28).…”

“…Particle size and high specific surface area provide suitable flow characteristics which are used for the purpose of improving post-dry properties (27). In this context, powders having different granulometries could be administered by means of a dry powder inhaler with an appropriate drug delivery along the respiratory tract (28). The behavior of post-dry redispersion is evaluated by the wet diffraction technique to demonstrate that the powders from a dispersion time are able to recover their nanometric characteristics (29).…”

“…A flow rate of 1.0 mL/min and injection volume 20 μl at ambient temperature were maintained while detection was performed at wavelength of 268 nm 41 . Prior to analysis, reverse phase column was equilibrated with mobile phase made up of methanol 5 mM sodium phosphate buffer in ratio of 595, and pH adjusted to 5 42 .…”

“…A wide variety of drug carriers have been developed to enhance the pharmacokinetic performance and biodistribution of drugs; however, the carrier is generally just an excipient for delivery goal where only the drug molecule is the therapeutically relevant compound . Natural compound‐based nanoparticles are a minimally explored area of drug delivery with significant promise for cancer therapy.…”
