An enhancer trap in the ascidian Ciona intestinalis identifies enhancers of its Musashi orthologous gene

Abstract: The enhancer trap technique, established in Drosophila melanogaster, is a very sophisticated tool. Despite its usefulness, however, there have been very few reports on enhancer traps in other animals. The ascidian Ciona intestinalis, a splendid experimental system for developmental biology, provides good material for developmental genetics. Recently, germline transgenesis of C. intestinalis has been achieved using the Tc1/mariner superfamily transposon Minos. During the course of that study, one Minos insertio… Show more

“…Microinjection-mediated enhancer detection is the simplest method (Sasakura et al, 2003;Awazu et al, 2004Awazu et al, , 2007. Its technical advantage is that we have only to prepare a transposon vector for enhancer detection to perform this method.…”

“…In fact, several Tc1/mariner transposons have shown their activity in both protostomes and deuterostomes (Loukeris et al, 1995a,b;Klinakis et al, 2000a, b;Shimizu et al, 2000;Zhang et al, 2002;Drabek et al, 2003;Pavlopoulos and Averof, 2005;Metaxakis et al, 2005). Genetic techniques such as germline transgenesis, enhancer detection (also called "enhancer trapping"), and insertional mutagenesis have been established in Ciona with Minos (Sasakura et al, 2003bAwazu et al, 2004).…”
